摘  要:目的探讨60Coγ射线大剂量辐射损伤对小鼠血液microRNA的影响及意义。方法 SPF级C57BL/6J小鼠接受10 Gy全身照射后,分别于照射后6、24 h进行外周血白细胞计数。同时应用Agilent microRNA生物芯片筛选小鼠血液中差异表达microRNA。结果 microRNA芯片筛选出辐射后6 h有27个差异表达的microRNA,其中12个上调,15个下调。辐射后24 h有26个差异表达的microRNA,其中4个上调,22个下调。与未照射组比较,表达差异有统计学意义(P<0.05)。10 Gy照射后,6和24 h外周血白细胞总数与对照组相比,差异有统计学意义。结论 microRNA在血浆中差异表达与辐射损伤有关,其有望成为辐射损伤新的血液标志物。[Objective]To investigate the influence and significance of high-dose 60Coγ radiation on serum microRNA in mice. [Methods]SPF C57BIJ6 J mice were treated with 10Gy whole body radiation, and the peripheral WBC counts were conducted at the 6th hour and the 24th hour after the treatment. Meanwhile, the differential expressions of of micro RNA were analyzed by Agilent miRNA microarrays. [Results]At the 6th hour after the radiation, 27 microRNA were differentially expressed, including 12 up-regulated microRNA and 15 down-regulated microRNA. At the 24th hour after the treatment, 26 microRNA were differentially expressed, including 4 up-regulated microRNA and 22 down-regulated microRNA. There was a statistically significant difference in microRNA expression between the radiation group and the non-radiation group (P〈0.05). The differences in total number of peripheral WBC at the 6th hour and the 24th hour after 10 Gy radiation were statistically significant between the radiation group and the control group. [Conclusion]The differential expression of serum microRNA is related to radiation damage, and it can be used as new biomarkers for exposure to high dose radiation.
