摘  要:目的研究慢性哮喘患者肺组织中IL-21和转录激活因子3(p-STAT3)表达水平。方法选择30只小鼠随机分为实验组和对照组,实验组进行哮喘干预,检测IL-21和p—STAT3的表达水平。实验组小鼠分别在第1、14天皮下注射0.15ml卵清白蛋白(ovalbumin,OVA)混合试剂,在第20天将小鼠放置于实验箱中,用雾化的方法将OVA混合试剂喷向实验箱中,2次/d,每次20min,观察小鼠出现的症状,对照组则在第1、14天皮下注射0.15ml无菌氯化钠注射液,其余处置同实验组小鼠。结果实验组小鼠的炎性细胞总数为(18.80±1.82)×10^5/L,嗜酸粒细胞数为(1.02±0.22)×10^5/L,单核细胞为(1.56±0.24)×10^5/L。与对照组比较,差异有统计学意义(t=2.449、2.656、2.361,P值均〈0.05)。实验组小鼠的IL=21表达水平为(65.45±5.56)ng/L,IL-2表达水平为(55.48±12.11)ng/L,p-STAT3表达水平为(76.86±9.46)ng/L,与对照组比较,差异有统计学意义(t=13.057、12.694、3.982,P值均〈O.05)。结论慢性哮喘小鼠肺组织IL=21和p-STAT3表达水平明显增加。Objective To study IL-21 and activating transcription factor 3 (p-STAT3) expression level in the lung tissue of patients with chronic asthma. Methods 30 mice were randomly divided into experimental group and control group, the experimental group of asthma intervention, the detection of IL- 21 and p-STAT3 expression. Mice in experimental group 1,14 days of subcutaneous injection of 0.15 ml ovalbumin (OVA) mixed reagent, on the twentieth day mice were placed in the experimental box, with the atomized method OVA mixed reagent is sprayed to the experimental box, two times a day, every 20 minutes, the mice were observed for symptoms, in 1,14 days of subcutaneous injection of 0.15 ml sterile sodium chloride injection group, with the rest of the disposal of experimental mice. Results The total number of inflammatory cells in mice of the experimental group is (18. 804-1.82)×10^5/L, eosinophil count is (1.02±0.22)×10^5 /L, mononuclear cells is (1.56±0.24)×10^5/L,compared with the control group, there were statistically significant ( t =2. 449,2. 656,2. 361, all P 〈0.05). The expression level of the mice of the experimental group IL-21 was (65.45 ± 5.56) ng/L, the expression level of IL-2 was (56.48± 12.11) ng/L, the expression level of p-STAT3 was (76.86±9.46) ng/L, compared with the control group, the difference was statistically significant ( t : 13. 057, 12. 694, 3. 982, all P 〈 0.05 ). Conclusions Chronic lung tissue of asthmatic mice and the expression level of p-STAT3 was significantly increased in IL-21.
