摘  要:为了研究高压蒸气灭菌对培养基pH值的改变及其对细菌生长的影响,试验采用临床或实验室常用的12种细菌培养基,通过检测其高压灭菌前后培养基pH值的变化,分析其各组成成分对pH值改变的影响,并选择在高压灭菌后pH值显著发生改变的THB培养基进行细菌的生长速率检测。结果表明:121℃高压蒸气灭菌25 min会使志贺氏菌增菌肉汤、葡萄糖肉浸液的pH值显著降低,使THB培养基的pH值显著升高(P<0.05),而对其他9种试验所用培养基不产生显著影响。说明高压蒸气灭菌对培养基pH值的改变是与培养基中的葡萄糖灭菌后可使溶液pH值降低及Na HCO3/Na_2HPO_4缓冲对灭菌后可使溶液的pH值显著升高有关。高压蒸气灭菌后不校正培养基的pH值会明显延长所培养菌株的静止期,同时还会使平台期的活菌总数减少。To study the effect of high- pressure steam sterilization on the changes in the pH value and bacterial growth,twelve kinds of commonly used bacterial culture media in the clinic and laboratory were used in the test. The effects of various components of these media on the changes in the pH value were analyzed through detecting the changes in the pH value in the culture media before and after high- pressure steam sterilization. THB culture medium that the pH value occurred significant changes after high- pressure steam sterilization was selected for a test of detecting bacterial growth rates. The results showed that 121 ℃ high- pressure steam sterilization for 25 min could significantly decrease( P 0. 05) the pH values in the Shigella enrichment broth and glucose meat infusion,and significantly increase the pH value in the THB culture medium( P 0. 05),while it had no significant effects in the other nine culture media used for the test. The results indicate that the glucose and the Na HCO3/ Na_2HPO_4 buffer pair were the main factors which affected the pH values of the media to be decreased or increased after high- pressure steam sterilization. The pH values in the media that are not corrected after high- pressure steam sterilization,can obviously prolong the stationary phase of the cultured strains,while also bringing the reduction of the total number of viable bacteria on the plateau phase.
