摘  要:目的优化南极放线菌XE-1发酵工艺,提高其抑菌活性物质产量并对其产生的活性代谢产物性质进行了初步研究。方法以发酵液中抑菌物质的活性为指标,用单因素实验和正交实验方法对放线菌菌株XE-1的发酵培养基和发酵条件进行优化;观察温度、pH值、储藏温度对抑菌物质稳定性的影响并进一步了解抑菌物质的极性大小。结果南极放线菌XE-1的最佳发酵培养基:可溶性淀粉30g/L,黄豆粉10g/L,海水晶35g/L,KON31.5g/L;最佳发酵条件:温度28℃,起始pH=8,接种量2%,摇床转速110r/min,发酵时间3d;抑菌物质热稳定性好,-20℃储藏活性基本不变,在强碱环境中易失活,不耐强碱,活性物质极性较小。结论确定了极地放线菌XE-1的最佳发酵培养基和最佳发酵条件,抑菌物质热稳定性好,在-20℃储藏活性基本不变,极性较小,不耐强碱。Objective To optimize the fermentation conditions with the aim of improving the antibacterial substance of Antarctic Streptomyces sp.XE-1,and to study the characteristics of active substances secreted by strain XE-1.Methods Single-factor experiment and orthogonal tests were used to optimize the fermentation medium and conditions.The effect of pH,temperature and storage temperature on the stability of active substances was investigated to study the hydrophobic nature of the antibacterial substances.Results The optimal fermentation medium:soluble starch 30g/L,soybean 10g/L,sea crystal35g/L,KNO31.5g/L;the optimal fermentation conditions:temperature 28℃,starting pH 8,inoculum concentration 2%,shaking speed 110r/min,fermentation time 3days.The active substances exhibited stability against heat and the activity was essentially the same under-20℃storage,but the active substance possessed less polarity and lost activity under strong alkali environment.Conclusion The optimal fermentation medium and conditions were obtained and the active substances possessed the great stability against heat and under-20℃storage,less polarity.It exhibited antibacterial activity at pH 2~12,whereas it was inactive under high alkalic condition.
