摘  要:目的探讨肚阿片受体基因(OPRM1)6个位点rs6912029、rs510769、rs3798676、rs7748401、rs589046和rs2075572单核苷酸多态性(Single nucleotide polymorphism,SNP)与海洛因成瘾者美沙酮维持治疗剂量的相关性。方法问卷调查收集696例美沙酮维持治疗门诊患者既往吸毒信息和目前治疗信息,结合多重PCR技术、MassARRAYiPLEX单碱基延伸技术和基质辅助激光解吸附电离飞行时间质谱分析技术对研究对象OPRM1基因6个位点进行SNP分型,探讨上述位点多态性与美沙酮维持治疗剂量间的关联。结果OPRM1基因rs6912029、rs510769、rs3798676、rs7748401、rs589046和rs2075572共6个位点的等位基因、基因型和单体型频率在美沙酮治疗高剂量和低剂量两组间分布差异均无统计学意义(P〉0.05)。结论尚未发现OPRM1基因6个SNP位点(rs6912029、rs510769、rs3798676、rs7748401、rs589046和rs2075572)与海洛因成瘾者美沙酮维持治疗剂量之间存在相关性。Objective To explore the association between the single nucleotide polymorphisms (SNPs) of OPRM1 (opioid receptor, mu 1 ) gene and methadone maintenance treatment (MMT) dose. Methods The genotypes of the six SNPs (rs6912029, rs510769, rs3798676, rs7748401, rs589046 and rs2075572) of OPRM1 gene in 696 patients from methadone maintenance treatment clinics were detected by multiple PCR assay, MassARRAY iPLEX single- base extension technology and matrix-assisted laser desorption/ionization time of flight (MALDI- TOF) SNP genotyping technique. The past drug use and treatment information were collected by questionnaires. The associations between MMT dose and the six SNPs were analyzed. Results The frequencies of genotypes, alleles and haplotypes of the six SNPs (rs6912029, rs510769, rs3798676, rs7748401, rs589046, rs2075572) were not significantly different between the high dose and low dose MMT groups(P 〉0.05). Conclu- sion No significant relevance is found between the OPRM1 gene SNPs (rs6912029 rs510769 rs3798676, rs7748401, rs589046, rs2075572) and methadone maintenance treatment dose in heroin abusers.
