摘 要:目的:观察大鼠脊髓腰段全横断损伤后beclin-1及脊髓神经元超微结构的变化。方法:雄性Wistar大鼠随机分成损伤后6h,1、3、7、14d和21d及假手术组共7组。脊髓损伤组用手术刀片横切腰段。电镜观察脊髓损伤以下神经元内超微结构形态学变化,免疫印迹检测beclin-1蛋白的表达。结果:脊髓损伤早期出现自噬体和自噬溶酶体,神经元内细胞器以退行性变化为主,随着时间延长,神经元超微结构出现明显改变;脊髓损伤后6h,1、3、7、14d和21d,与假手术组相比,大鼠脊髓组织中beclin-1蛋白明显增加,差异有统计学意义;脊髓损伤后3dbeclin-1蛋白表达最高,7d开始降低,21d降到最低。结论:脊髓腰段全横断损伤后神经元自噬活动增强,beclin-1可反映神经元自噬状态的变化。Objective: To explore the effects of lumbar spinal cord injury on expression of beclin-1 and changes of ultrastructure of neurons in rats with completely transected spinal cord. Methods: Male Wistar rats were randomly divided into 7 groups: 6 h, 1 d, 3 d, 7 d, 14 d and 21 d after spinal cord injury and a sham-operated group. In the spinal injury group, the models were built by cutting off lumbral segment with surgery blade. Morphological changes of various organelles of neurons below the spinal cord injury were observed by electron microscopy. The expression of beclin-1 was examined by Western blotting. Results: The formation of autophagosome and autolysosome was investigated under electronic microscope and some organdies of neurons were degenerated at the early stage after spinal cord injury. The ultrastructure changed obviously with the extension of the time Compared with the sham- operated group, the expression of beclin-1 in the injury group was increased significantly. The expression of beclin-1 was enhanced in 3 d group after injury than that of the other groups. However, after injury, the expression began to decrease in 7 d group and reached a minimum in 21 d group. Conclusion: Transection injury of lumbal segments of spinal cord induces the increase of autophagy in rats. Beclin-1 may be involved in the process of autophagy in neurons.
