摘 要:目的研究大鼠脑震荡伤后脑组织中诱导型一氧化氮合酶(i NOS)与胶质纤维酸性蛋白(GFAP)表达比值的变化规律及其与脑震荡损伤时间之间的关系,以期为脑震荡的损伤时间推断提供新的思路。方法将85只健康Sprague-Dawley大鼠随机分成模型组(25只)、实验组(55只)及对照组(5只),模型组大鼠用于确定打击高度以建立脑震荡模型;实验组采用自由落体法建立脑震荡模型后根据处死的时间不同再分为0.5、1、3、6、12、24、48、96、168、240、336 h共11组,每组5只;对照组大鼠于常规饲养24 h后处死。实验组及对照组大鼠取脑组织作常规苏木精-伊红染色及GFAP和i NOS免疫组织化学染色,对结果进行图像分析。结果伤后0.5~3 h,i NOS与GFAP阳性表达的比值由5.03上升至10.47,呈逐渐升高的趋势;此时i NOS阳性表达亦显著升高,阳性细胞率由14.61%上升至37.45%,但GFAP的阳性表达无明显升高。伤后3~12 h,二者的比值逐渐降低至4.98,但仍然处于较高水平;同时i NOS阳性表达与GFAP的阳性表达均显著升高。伤后12~336 h,二者比值呈单调递减的变化趋势,由4.98降低至0.95,且均小于0.5~12 h的二者比值;同时i NOS和GFAP的阳性表达均表现为持续升高至峰值后再降低的变化趋势。结论采用i NOS与GFAP比值,同时结合i NOS与GFAP各自变化规律,可为脑震荡伤损伤时间的推断提供依据。两个或多个生物化学指标的比值研究,可望为损伤时间的推断提供可行的研究思路。Objective To study the relationship between the expression ratio of induced nitric oxide synthase(i NOS) over glial i brillary acidic protein(GFAP) and the time of injury at er brain concussion in rat,in order to acquire a new visual angle for determining injury time of cerebral concussion.Methods Eighty-i ve healthy Sprague-Dawley rats were divided into three groups randomly: model group(n=25),experimental group(n=55),and control group(n=5).The rats in the model group were used to coni rm the attack hight to make the model of brain concussion; according to the time of execution,rats in the experimental group were then subdivided into 11 groups with 5 rats in each subgroup,and their execution time was respectively hour 0.5,1,3,6,12,24,48,96,168,240,and 336; the rats in the control group were executed at er fed for 24 hours.At er the model of cerebral concussion was established through freefalling dart method,hematoxylin-eosin staining and immunohistochemistry staining of i NOS and GFAP were conducted for the brain of the rats.All related experimental results were studied by using microscope with image analytical system and homologous statistics.Results The ratio of positive expression of i NOS over that of GFAP increased gradually during hour 0.5-3 at er injury in brain(from 5.03 to 10.47).At the same time,the positive expression of i NOS increased signii cantly(from 14.61% to 37.45%).However,the increase of the positive expression of GFAP was not obvious.Between hour 3 and 12,the ratio began to decline to 4.98,which was still at a high level,and during the same time period,the positive expressions of i NOS and GFAP also experienced the same change pattern.Later,the ratio began to decline between hour 12 and 336 at er injury(from 4.98 to 0.95).All ratios at this time were lower than those between hour 0.5 and 12.h e positive expression of i NOS and GFAP both increased to a climax before declining.Conclusions h e ratio of positive expression of i NOS over GFAP and the respective c
