黄芪保心汤对扩张型心肌病大鼠心室重构的影响

摘　　要：目的观察黄芪保心汤对盐酸阿霉素致扩张型心肌病(DCM)大鼠心功能及心室重构的影响。方法55只大鼠分为正常组6只和造模组49只,造模组使用阿霉素腹腔注射8周后,36只DCM成模大鼠分为:中药组给予黄芪保心汤5 m L/kg灌胃,西药组予卡托普利50 mg/kg,联合组予卡托普利和黄芪保心汤联合给药,造模组作为模型组不予处理,对照组和造模组正常喂饲。4周后再次行心脏彩超观测心室重构的变化,酶联免疫法(ELISA)检测血清脑钠素(BNP)、基质裂解素2(ST2)的含量,测定左室质量分数(LVMI),苏木素-伊红(HE)染色观察心肌形态,天狼猩红苦味酸(SR)染色测定心肌胶原容积分数(CVF)。结果与造模组比较,中药组、西药组、联合组左室舒张末期内径(LVEDD)、左室收缩末期内径(LVESD)、左心室射血分数(EF)、左心室短轴缩短率(FS)均有显著差异(P<0.05),BNP、ST2水平均有降低(P<0.01),LVMI及CVF有显著降低(P<0.01)。结论黄芪保心汤可以改善扩张型心肌病大鼠心功能及延缓心室重构进程,联合西药效果更加显著。Objective：To observe the influence of Huangqi Baoxin（HQBX）decoction on heart functions and cardiac fibrosis in rats with dilated cardiomyopathy（DCM）.Methods：Fifty-five rats were divided into the normal group（6）and DCM model group（49）in which rats were given intraperitoneal injection of doxorubicin for eight weeks.Echocardiography was made to choose the DCM rats which were then randomly divided into group B,C,D and E.Group C were given HQBX decoction 5 m L/（kg·d）;group D received captopril 50 mg/（kg·d）;group E adopted a combination of HQBX decoction and captopril with the same volume mentioned.Echocardiography was made again to explore heart functions including LVEDD,LVESD,EF and FS.Left ventricular mass index（LVMI）was also explored.ELISA（enzyme-linked immunosorbent assay）was used to assess the levels of BNP,ST2,then HE staining to detect myocardium shape,and SR staining to differenciate collegen volume fraction（CVF）.Results：Compared with group B,LVEDD,LVESD,EF and FS changed remarkably（P〈0.05）;BNP and ST2 greatly reduced（P〈0.01）;LVMI and CVF both notably decreased（P〈0.01）.Conclusions：HQBX decoction can improve heart function and ventricular remodeling in rats with dilated cardiomyopathy,and make a better effect plus western medicine.
