益气养阴通络法对兔腹主动脉内膜剥脱术后内皮功能的影响

摘　　要：目的探讨具有益气养阴通络功效的通脉颗粒防治经皮冠状动脉介入治疗(PCI)术后再狭窄的可行性。方法将48只新西兰大耳兔按照随机数字表法分为正常组、模型组、通脉颗粒组(中药组)、阿托伐他汀组(西药组),各12只。正常组未经手术及用药,模型组、中药组、西药组用球囊导管剥脱兔腹主动脉内膜行手术造模。模型组术前及术后未用药;中药组于术前1 d开始给予通脉颗粒灌胃给药,连续用药3周;西药组于术前1 d开始给予阿托伐他汀口服,连续用药3周。3周后采血检测内皮素(ET-1),一氧化氮(NO);Western方法检测兔腹主动脉血管内皮生长因子(VEGF)、成纤维细胞生长因子(b FGF)、转化生长因子(TGF)-β1蛋白表达。结果中药组和西药组可明显增加血浆NO,降低ET-1;中药组、西药组可增加VEGF蛋白表达,减少b FGF、TGF-β1蛋白表达(均P<0.05)。结论通脉颗粒能够促进损伤血管内皮细胞生长及内膜修复,其机制可能与降低血浆ET-1,升高血浆NO,增加VEGF,减少b FGF、TGF-β1蛋白表达有关,初步具有防治PIC术后再狭窄的作用。Objective：To investigate the feasibility of Yiqi Yangyin Tongluo of Tongmai particles in Prevention efficacy of restenosis after Percutaneous coronary intervention（PCI）.Methods：48 New Zealand rabbits were randomly divided into four groups,the normal group,the model group,Tongmai Particles group（TCM group）,and atorvastatin group（western medicine group）,12 in each.The normal group received no surgery and medicine,while the other groups were modeled with balloon catheter denudation rabbit abdominal aorta surgery.The model group remained untreated before and after surgery;a day before surgery,TCM group were administered with Tongmai Particles,continuous medication for 3 weeks,while western medicine group took atorvastatin orally,continuous medication for 3 weeks,too.After three weeks,blood was taken to test endothelin（ET-1）and nitric oxide（NO）.Protein expression of VEGF,b FGF TGFβ-1 of abdominal aorta in rabbit was detected with Western method.Results：NO rose and ET-1 reduced in plasma in both TCM group and western medicine group;in Chinese medicine group and western medicine group,the protein expression of VEGF could increase and the protein expression of b FGF and TGFβ-1 could reduce（P〈0.05）.Conclusion：Tongmai Particles can promote growth and endometrial repair of vascular endothelial cell,which may be associated with the decrease of plasma ET-1,rise of plasma NO,increase of VEGF and reduce of protein expression of b FGF and TGFβ-1,initially play a role in Prevention of surgery PIC restenosis.
