摘 要:目的探讨丹参酮ⅡA治疗大鼠子宫内膜异位症(EM)的作用机制。方法 SD大鼠72只,按照随机数字表法分为对照组,模型组,达那唑组,丹参酮小、中、大剂量组。将大鼠自体子宫内膜组织经皮下移植于模型组、达那唑组及丹参酮ⅡA小、中、大剂量组右侧腹部,制作大鼠EM动物模型。造模后丹参酮小、中、大剂量组用丹参酮ⅡA灌胃治疗14 d,达那唑组灌达那唑,对照组及模型组灌等体积0.9%氯化钠注射液。测定大鼠尾静脉血清雌二醇(E2)、白介素-2(IL-2),免疫组织化学法检测子宫内膜组织雌激素受体(ERα)及细胞增殖核抗原(PCNA)表达量。采用ELISA法测定大鼠子宫内膜组织VEGF蛋白及VEGF m RNA,低氧诱导因子-1α(HIF-1α)、HIF-1αm RNA等表达量。结果与模型组比较,丹参酮ⅡA中、大剂量组血清IL-2、E2均降低,异位子宫内膜组织ERα、PCNA、VEGF蛋白及VEGF m RNA表达量表达均下调(P<0.05)。结论丹参酮ⅡA抑制大鼠子宫异位内膜生长的作用机制可能与其祛瘀、活血调经的药性及下调异位子宫内膜组织上VEGF蛋白及VEGF m RNA表达,抑制移植的子宫内膜组织血管新生,降低IL-2、E2分泌,减轻血管内皮炎症反应造成的组织纤维化增生有关。Objective:To discuss the mechanism of Tanshinone ⅡA on endometriosis in rats.Methods:72 SD rats were randomly divided into the control group,the model group,danazol group,Tanshinone Ⅱ A groups(low,mid,high).Endometrial tissue was transplated subcutaneouly in right abdomen in the model group,danazol group,and Tanshinone Ⅱ A groups(low,mid,high)to make EM rat models.After modeling,Tanshinone Ⅱ A groups took intragastric administration of Tanshinone ⅡA for 14 days,while danazol group,the control group and the model group tooke the same volume of 0.9% sodium chloride injection.E2 and IL-2 in caudal vein serum of rat were determined.The expression of estrogen receptor(ER)and proliferating cell nuclear antigen(PCNA)in endometrial tissue was detected by immunohistochemistry.ELISA method was used to measure the expression levels of VEGF protein and VEGF-m RNA,hypoxia inducible factor-1 alpha(HIF-1 alpha),HIF-1 alpha m RNA in rat endometrial tissue.Results:Compared with the model group,both IL-2 and E2 in serum dropped,as well as the expression of ERα,PCNA,VEGF and VEGF-m RNA in ectopic endometrial tissue in Tanshinone ⅡA groups(mid,high)(P〈0.05).Conclusion:The mechanism of Tanshinone ⅡA on inhibiting the growth of Ectopic endometrium may ralted to its drug properties in removing blood stasis and anti-inflammatory,promoting blood flow to regulate menstruation,decreasing the expression of VEGF and VEGF-m RNA in ectopic endometrial tissue,inhibition on Angiogenesis of transplanted endometrial tissue,decreasing secretion of IL-2,E2,and relieving tissue fibrosis caused by vascular endothelial inflammatory response.
