姜黄素对脓毒症大鼠小肠功能保护作用的研究

摘　　要：目的探索姜黄素对脓毒症大鼠小肠功能的保护作用及其机制。方法 30只SD大鼠,按照随机数字表法分为假手术组、脓毒症组和姜黄素治疗组,每组10只。采用改良盲肠结扎穿孔方法建立大鼠脓毒症模型,假手术组除不结扎刺穿盲肠外,其余操作同脓毒症组,姜黄素治疗组模型建立后1 h腹腔注射姜黄素(二甲亚砜作为溶剂),假手术组及脓毒症组注射同样剂量的二甲亚砜。于手术后24 h、48 h处死大鼠,测定小肠黏膜组织的含水量;小肠黏膜组织中三磷酸腺苷(ATP)、二磷酸腺苷(ADP)、单磷酸腺苷(AMP)含量;观察并比较各组小肠黏膜上皮组织病理改变。结果姜黄素治疗组的小肠组织水肿程度较假手术组明显升高(P<0.05),较脓毒症组明显降低(P<0.05);姜黄素治疗组的小肠黏膜ATP、ADP含量较脓毒症组均有明显升高(P<0.05)。AMP含量明显下降(P<0.05);小肠黏膜上皮组织病理学提示姜黄素治疗组的组织损伤明显轻于脓毒症组。结论姜黄素对脓毒症大鼠小肠组织水肿和能量代谢的抑制状态具有一定程度的改善作用。Objective：To study the protective effect and mechanism of curcumin on small intestinal function in septic rats.Methods：30 SD rats were randomly divided into the sham group,sepsis group,and curcumin group.Septic models were induced with cecal ligation and puncture（CLP）.In the sham group,the abdomen was opened without CLP.In curcumin group,curcumin was intravenously injected curcumin in abdomina after CLP.Five rats of the each group were sacrificed at 24 h and 48 h respectively after CLP.Moisture capacity levels of ADP,ATP and AMP of the small intestinal mucosa were measured.Pathological changes of small intestinal mucosa were observed.Results：The levels of tissue edema in the small intestinal mucosa of curcumin group were significantly higher than those of the sham group（P〈0.05）,while significantly lower than the sepsis group（P〈0.05）.Compared with the septic group,the levels of ATP and ADP in the small intestinal mucosa of curcumin group were significantly higher（P〈0.05）;and the level of AMP of the small intestinal mucosa of curcumin group was much lower（P〈0.05）;severer pathological changes were found in the sepsis group than that in curcumin group.Conclusion：In some degree,curcumin may improve edema of small intestine and Inhibition of energy metabolism in rats with sepsis.
