卡巴匹林钙在猪体内的药代动力学和安全性研究

摘　　要：研究了猪单剂量口服、静注40 mg/kg.bw卡巴匹林钙后其代谢产物阿司匹林和水杨酸的药代动力学过程,并按照1、3、5倍推荐剂量进行了卡巴匹林钙可溶性粉对靶动物猪的安全性试验。药动学试验通过HPLC-UV测定不同采样时间点的血药浓度,数据用Win Nonlin5.2.1软件的非房室模型进行拟合。结果表明,卡巴匹林钙给药后,代谢产物阿司匹林和水杨酸达峰快,峰浓度高。安全性评价试验则通过测定血常规、血生化指标来评估。给药组与对照组相比,WBC、RBC、HGB等血液生理常规指标及AST、GLU、TG、TC等血液生化指标没有显著差异,临床也未观察到不良反应。结果表明,卡巴匹林钙可溶性粉5倍推荐剂量给药,对靶动物猪是安全的。The pharmacokinetics of aspirin and salicylic acid,carbasalate calcium＇s metabolites were investigated in pig by oral administration and intravenous injection at a single dosage of 40 mg/kg body weight. The safety of carbasalate calcium was evaluated by oral administration at 1,3,5 times of the recommended dose of carbasalate calcium soluble powder. In the pharmacokinetic study,the plasma samples collected at different points of time were determined by HPLC-UV. The data were analyzed by noncompartmental model of Win Nonlin5.2.1 software. The results showed that,time to peak of aspirin and salicylic acid was short and peak concentrations of them were high. Safety evaluation of carbasalate calcium was carried out by detecting hematological parameters,biochemical indicators. Comparing the test group and control group,there was no significant difference between them in regular blood physiology indicators like WBC,RBC,HGB,etc. and serum biochemistry indicators like AST,GLU,TG,TC,etc. There was no clinical adverse reaction. The results indicated that the carbasalate calcium soluble powder was safe to pig at the dosage of 5 times of recommended dose.
