摘 要:目的比较漏斗冷冻法和直接浸入冷冻法大鼠脑组织提取物的磁共振波谱(MRS),为进一步研究提供依据。方法成年健康SD大鼠随机分为两组,分别用漏斗冷冻法和直接浸入冷冻法取脑组织,制备高氯酸组织提取物,溶于重水(D_2O)分别用31PMRS和1H MRS检测。结果 31P MRS可以检测出多种含磷代谢产物,脑内PCr/Pi、ATP/Pi、ATP/EPP三组比值漏斗冷冻法明显高于直接浸入法;1H MRS可检测出N-乙酰天冬氨酸(NAA)、乳酸、肌酸及多种氨基酸,漏斗冷冻法与直接浸入法相比,NAA含量高,乳酸含量低。结论漏斗冷冻法可以较好保存脑内的能量代谢产物,脑组织提取物MRS可以检测出多种含磷和含氢代谢产物,二者结合是比较好的脑代谢研究方法。Objective To compare magnetic resonance spectroscopy of brain tissue extracts after funnel freezing and direct dipping freezing.Methods Twelve adult SD rats were divided into two groups,one frozed with funnel and another frozed by direct dipping into liquid N2.The brain tissue extracts were dissolved in 0.5mL D2O for 31P and 1H MRS analysis.Results 31P MRS could reveal many phosphrus-containing metabolites of brain.The ratio PCr/Pi,ATP/Pi,ATP/EPP in funnel freezing group were higher than that in direct dipping freezing group.1H MRS could display N-acetylaspartate,lactic acid,creatine and many amino acids.There was higher content of N-acetylaspartate and lower content of lactic acid contained in funnel freezing group than in direct dipping freezing group.Conclusions Funnel freezing method could comparatively preserve capacity metabolites.31P and 1H MRS may reveal many metabolites of brain.Integrating two methods together was a relatively perfect research way about capacity metabolism of brain.
