摘  要:目的探讨慢性饮酒及戒断对大鼠学习记忆及海马PSD-95表达的影响。方法 36只成年雄性SD大鼠随机分为对照组、乙醇组和戒断7 d组,对照组正常饮水饲养,乙醇组以6%(v/v)的酒精作为大鼠唯一饮水来源,持续30 d,戒断7 d组以6%(v/v)的酒精作为大鼠唯一饮水来源,持续30 d后,恢复正常饮水7 d。戒断症状评分评价大鼠的酒精戒断行为体征,Morris水迷宫实验观察不同组别大鼠学习记忆成绩,免疫组化和Western blot检测不同组别大鼠海马PSD-95蛋白表达的变化。结果柳田知司评分显示:乙醇组显著性高于对照组(q=22.31,P<0.01),戒断7 d组显著性低于乙醇组(q=19.19,P<0.01)。Morris水迷宫实验结果显示:与对照组比较,乙醇组大鼠逃避平台潜伏期时间连续4个实验日均显著延长(qd1=5.87,qd2=6.61,qd3=7.54,qd4=6.43,P均<0.01),目标象限停留显著变短(q=17.15,P<0.01),穿越平台明显减少(q=7.35,P<0.01);与乙醇组比较,戒断7 d组大鼠逃避平台潜伏期时间连续4个实验日均显著变短(qd1=4.12,qd2=3.92,qd3=5.37,qd4=4.38,P均<0.01),目标象限停留显著延长(q=7.83,P<0.01),穿越水下平台明显增加(q=3.55,P<0.05)。免疫组化和Western blot结果均显示:与对照组比较,乙醇组海马PSD-95的表达显著减少(q=16.49,P<0.01),与乙醇组比较,戒断7 d组海马CA1区PSD-95的表达显著升高(q=6.98,P<0.01)。结论酒精依赖可使大鼠的学习记忆能力减退,实施戒断之后学习记忆能力可以得到改善,可能与海马PSD-95蛋白的表达变化有关。Objective This work was to investigate the effect of chronic alcohol consumption and withdrawal on the learning learning and memory,and PSD- 95 Expression and PSD- 95 Expression in hippocampus of rats. Methods 36 adult male SD rats were randomly divided into three groups: a control group,an ethanol group and a 7- day withdrawal group,ethanol group and 7- day withdrawal group. The control group was fed with normal ater,the ethanol group was fed with 6%( v/v) ethanol as the sole source of water for 30 days. The 7- day withdrawal group was fed with 6%( v/v) ethanol as the sole water source for 30 days,and then fed with normal water for 7 days. The alcohol withdrawal signs was evaluated by using withdrawal symptom score. The learning and memory was observed by morris water maze test,and the changes in PSD- 95 protein expression in hippocampal were tested by immunohistochemistry and Western blot. Results The withdrawal scores of the ethanol group was significantly higher than the control group( q = 22. 31,P〈0. 01). That of the 7- day withdrawal group was significantly lower than ethanol group( q = 19. 19,P〈0. 01). The morris water maze results showed that compared with the control group,the escaping latency time of the ethanol group was significantly longer( qd1 = 5. 87,qd2 = 6. 61,qd3 = 7. 54,qd4= 6. 43,P〈0. 01) for 4 experimental days,and the target quadrant residence time was significantly shorter( q = 17. 15,P〈0. 01),and the number of times across the platform was significantly reduced as well( q = 7. 35,P〈0. 01). Compared with the ethanol group,the escaping latency time of the withdrawal 7 days group was significantly shorter( qd1 = 4. 12,qd2 = 3. 92,qd3 = 5. 37,qd4 = 4. 38,P 均 0. 01,P〈0. 01) for 4 experimental days; the target quadrant residence time was significantly longer( q = 7. 83,P〈0. 01); the number of times across the platform increased significantly( q = 3. 55,P〈0. 05).
