摘 要:目的利用PCR array技术观测高脂血症大鼠肝脏脂蛋白和胆固醇代谢信号通路相关基因表达。方法 SPF级健康雄性SD大鼠20只,随机分为空白对照组、高脂血症组。使用全自动生化分析仪检测血清胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)水平;提取各组大鼠肝脏总RNA,应用脂蛋白和胆固醇代谢信号通路PCR芯片检测高脂血症大鼠肝脏脂蛋白和胆固醇代谢信号通路相关基因表达。结果与空白对照组相比,高脂血症大鼠血清HDL-C水平显著降低,TC、LDL-C水平显著升高,高脂血症大鼠上调≥2倍的基因6个,占7%,如Lrp1b、Stab2、Apol2、Abcg1、RGD1564999等基因;下调≥2倍的基因36个,占43%,如Ldlr、Lrp6、Pcsk9、Apoa4、Cdh13、Sorl1、Apoa1等,上述上调或者下调的基因,涉及脂蛋白信号通路的LDL受体、LDL受体相关蛋白、LDL相关蛋白及HDL相关蛋白相关基因;胆固醇转运途径的胆固醇转运、胆固醇流出、胆固醇逆向转运相关基因;以及胆固醇代谢途径的胆固醇吸收、分解代谢、胆固醇的平衡及生物合成等相关基因。结论高脂血症引起血清胆固醇水平异常,可能与调控肝脏的脂蛋白和胆固醇代谢信号通路相关基因表达改变密切相关。Objective To observe the related gene expressions of lipoprotein and cholesterol metabolism signaling pathways in hyperlipidemia rats by PCR array technology. Methods 20 SD rats were randomly divided into the control group and the HLP group. The rats in the HLP group were given high fat diet. The levels of TG,TC,HDL- C and LDL- C in serum were detected by automatic biochemical analyzer,the RNA was extracted from two groups,and PCR array was used to test the related gene expressions of lipoprotein and cholesterol metabolism signaling pathways in the rats' livers. Results Compared with the control group,the levels of TC and LDL- C increased while the level of HDL- C significantly decreased in the HLP group. PCR array showed that there were 6 gene expressions( 7%) increased by twice or more than twice( e. g. Lrp1 b,Stab2,Apol2,Abcg1,RGD1564999,Lep),36 gene expressions( 43%) decreased by twice or more than twice( e. g. Ldlr,Lrp6,Pcsk9,Apoa4,Cdh13,Sorl1,Apoa1,etc.). The above genes involved LLD receptors of Liporotein Signaling,relevant protein of LDL receptor,and relevant genes relating to protein involved in LDL and HDL,the related genes in cholesterol transport pathway include cholesterol transport,cholesterol efflux and reverse cholesterol transport,the genes relating to cholesterol metabolism pathways include cholesterol absorption,catabolism,cholesterol balance and biosynthesis and so on. Conclusion Abnormal serum cholesterol levels caused by hyperlipidemia may relate to the changes of gene expression which regulate hepatic lipoprotein and cholesterol metabolism signaling pathways.
