摘 要:目的观察血府逐瘀汤对心肌梗死大鼠心肌发育相关基因GATA4、NKx2.5的调控作用。方法采用结扎大鼠左冠状动脉前降支方法建立心肌梗死模型,将成模大鼠随机分为模型组和血府逐瘀汤组,每组30只。另选择同月龄健康SD大鼠10只作为假手术组。模型组给予生理盐水2 m L/次灌胃,血府逐瘀汤组给予浓煎血府逐瘀汤2 m L/次灌胃,均1次/d。采用RT-PCR方法检测3组大鼠干预1 d、10 d、15 d心肌组织中GATA4、Nkx2.5mRNA表达水平。结果干预1d后,模型组GATA4、Nkx2.5mRNA表达量均明显低于假手术组(P均<0.05);干预10 d后,模型组和血府逐瘀汤组GATA4、Nkx2.5mRNA表达量与假手术组比较差异均无统计学意义(P均>0.05),但模型组GATA4、Nkx2.5mRNA表达量和血府逐瘀汤组GATA4mRNA表达量均明显高于干预1 d后(P均<0.05);干预15 d后,血府逐瘀汤组GATA4、Nkx2.5mRNA表达量均明显高于假手术组(P均<0.05)。干预1 d、10 d、15 d后,血府逐瘀汤组GATA4、Nkx2.5mRNA表达量均较模型组有所升高,但差异均无统计学意义(P均>0.05)。结论大鼠心肌梗死后,其心肌组织中GATA4、Nkx2.5mRNA表达量呈现先下降、再升高的趋势。血府逐瘀汤可发挥一定的促进心肌梗死大鼠心肌发育相关基因GATA4、NKx2.5表达增加的作用,该作用可能是祛瘀中药促进梗死后心肌组织迅速修复、愈合的机制之一。Objective It is to explore the regulatory effect of Xuefu Zhuyu decoction on gene GATA4 and NKx2. 5 in rats after myocardial infarction. Methods The left anterior descending coronary arteries of SD rats were ligated to establish rat models of myocardial infarction. All rats with myocardial infarction were divided into model group( n = 30) and Xuefu Zhuyu decoction group( n = 30). Anther 10 rats were selected as sham group( n = 10). The rats in control group were treated with 2m L of 0. 9% Na Cl per day. The rats in Xuefu Zhuyu decoction group were treated with 2 m L of concentrated decoction per day.Treatment duration was also altered. There were three different duration of treatments specifically 1 day,10 days,and 15 days. The levels of GATA4,Nkx2. 5mRNA in myocardium were detected by RT-PCR. Results On the 1st day after myocardial infarction,the expression of GATA4 mRNA and Nkx2. 5mRNA in myocardial infarction model group were significantly decreased compared to those in the sham group( P0. 05). On the 10 th day after myocardial infarction,the expression of GATA4 mRNA and Nkx2. 5mRNA in model group and Xuefu Zhuyu decoction group were not significantly increased compared to those in the sham group( P0. 05),but the expression of GATA4 mRNA and Nkx2. 5mRNA in model group and the expression of GATA4 mRNA in Xuefu Zhuyu decoction group on the 10 th day were significantly increased compared to those in the sham group on the 1st day( P0. 05). On the 15 th day after myocardial infarction,the expression of GATA4 mRNA and Nkx2.5mRNA in Xuefu Zhuyu decoction group were significantly increased compared to those in the sham group( P0. 05). On the1 st,10th and 15 th day after myocardial infarction,the expression of GATA4 mRNA and Nkx2. 5mRNA in Xuefu Zhuyu decoction group were increased compared to those in the myocardial infarction model group,but there was not significant difference between the two groups( P 0. 05). Conclusion Xuefu Zhuyu decoction may have a regulatory effect on gene GATA4
