三华李成熟胚培养及子叶离体再生研究

摘  要:对影响三华李成熟胚萌发成苗及子叶离体再生的因素进行了研究。结果表明:种皮是抑制三华李萌发的主要因素,去除种皮就能促进三华李成熟胚的萌发。其中,以MS+6-BA0.5mg/L较为适宜三华李成熟胚离体萌发成苗。子叶不定茅的诱导以6-BA浓度0.5-1.0mg/L为好,不定芽继代与增殖较适宜培养基配方为F14+AgNO35mg/L+CH500mg/L+琼脂7g/L+蔗糖3%+0.5mg/L6-BA+0.5mg/LIBA,IBA最适合的不定根诱导浓度为9.0mg/L。The factors affecting the growth from mature embryos germination to seedling and cotyledon in vitro regeneration of Prunus salicina cv. sanhua are studied. Results show that the seed coat is the main factor for restraining germination of Prunus salicina cv. sanhua. The removal of the seed coat can promote the germination of mature embryos of Prunus salicina cv. sanhua. MS culture medium+ 0.5mg/L 6-BA concentration were suitable for the growth from germination of mature embryos of Prunus salicina cv. sanhua to seedling. Vitro. 1.0mg/L 6- BA is better for Cotyledon adventitious bud induction. The medium of F14+ AgNO3 5mg/L + 5mg/L + CH 500mg/ L + AGAR 7g/L + sucrose 30g/L + 6-BA 0.5mg/L + IBA 0.5mg/L is suitable for adventitious buds proliferation, and the IBA with 9.0mg/L concentration is most suitable for adventitious roots induction.
