摘  要:运用Box-Behnken响应曲面法设计料液比、提取次数、H2SO4浓度及提取时间4因素3水平的试验模型,以盐酸小檗碱的得率为评价指标,对黄连饮片中盐酸小檗碱的提取工艺进行优化。优选出最佳提取工艺参数为:H2SO4浓度为3%,料液比为1∶10,提取3次,每次2 h。测定盐酸小檗碱的得率为8.36%,与预测值8.30%基本相符合,偏差较小。该工艺稳定,可靠,为黄连饮片中盐酸小檗碱的研究提供了依据。To optimize the conditions for purification of berberine hydrochloride in Coptis chinensis Franch,Box-behnkn response surface method was used to design the test modls with four factors including liquid ratio,extraction times,H2SO4 concentration and extraction time at three levels based on the single factor experiment,and the pureness of berberine hydrochloride was used as the evaluation index,to optimize purification process of berberine hydrochloride. The optimal purification conditions were determined as extracted three times for 2 h,at a solid- liquid ratio was 1∶ 10,and the H2SO4 concentration was 3%. The determination of berberine hydrochloride at a rate of 8. 36%,and the predictive value of 8. 30% conforms to the basisi deviation is smaller. The process is stable and reliable,and the results of the study can provide scientific basis for the determination of the purification process of extraction of berberine hydrochloride in Coptis chinensis Franch.
