摘 要:目的探讨基于Actinomyces ruminicola的微生态制剂对瘤胃发酵的影响。方法将分离得到的能生成挥发性脂肪酸(volatile fatty acid,VFA)的瘤胃厌氧菌Actinomyces ruminicola(命名为NY2,以下简写为Ar)与痤疮丙酸杆菌(以下简写为Pa)按3种比例混合制成微生态制剂(M1:Ar∶Pa=2∶3;M2:Ar∶Pa=1∶1;M3:Ar∶Pa=3∶2),进行羊体内发酵试验,分别于饲喂后0、2、4、6、8、10、12和24 h无菌采集各组羊的瘤胃液,用精密p H检测仪测定瘤胃液的酸碱度;Waters2695液相色谱仪测定瘤胃液中VFA(主要是乙酸、丙酸和丁酸)的含量。结果各组瘤胃液的p H值均呈先下降后升高的趋势;M2组瘤胃液的p H值与M1和M3组差异有统计学意义(P均〈0.05)。当微生态制剂中Ar∶Pa=1∶1,饲喂4-6 h后,瘤胃液中乙酸、丙酸、丁酸和总VFA的浓度均达最高,分别为(82.49±2.51)、(35.12±1.69)、(22.92±1.02)和(139.81±5.61)mmol/L,该比例的微生态制剂可保持瘤胃内容物p H在6.0-7.0之间波动。结论本实验制备的微生态制剂有利于瘤胃微生物发酵,能促进乙酸、丙酸等VFA的生成,为今后该瘤胃微生态制剂的应用奠定了基础。Objective To investigate the effect of Actinomyces ruminicola(Ar, named as NY2)-based microecological preparation on rumen fermentation. Methods The isolated NY2 producing volatile fatty acid(VFA) was mixed with Propionibacterium acnes at ratios of 2 ∶ 3, 1 ∶ 1 and 3 ∶ 2 to prepare microecological preparations M1, M2 and M3,respectively, which were subjected to fermentation test in vivo in sheep. The rumen liquids of sheep in various groups were collected 0, 2, 4, 6, 8, 10, 12 and 14 h after feeding, and determined for p H value by p H meter, and for VFA(mainly acetic acid, propionic acid and butanoic acid) content by Waters2695 liquid phase chromatography. Results The p H values of rumen liquids in various groups decreased at first then increased. However, the p H value in M2 group showed significant difference than those in M1 and M3 groups(each P〈0. 05). The levels of acetic acid, propionic acid, butyric acid and total VFA in rumen fluids of sheep 4 - 6 h after being fed with M2 reached the maximums, which were(82. 49 ±2. 51),(35. 12 ± 1. 69),(22. 92 ± 1. 02)and(139. 81 ± 5. 61)mmol / L respectively. The p H value of rumen liquid of sheep fed with M2 fluctuated between 6. 0 and 7. 0. Conclusion The prepared microecologics were beneficial to the rumen microbial fermentation, and promoted the production of acetic acid, propionic acid and other VFAs, which laid a foundation of their application in future.
