摘 要:目的探讨隔药灸对溃疡性结肠炎大鼠(UC)血清Th17主要相关细胞因子的调控机制。方法将SD大鼠随机分为正常组、模型组、隔药灸组和西药组4组,采用免疫法加局部刺激造模,用ELISA法检测大鼠血清IL-17、IL-23、IL-6、TGF-β、IL-1β和TNF-α的表达水平。结果 UC大鼠血清IL-17、IL-23、IL-6、TNF-α、IL-1β水平均高于正常大鼠(P<0.05),而TGF-β低于正常组(P<0.05)。经过治疗后,隔药灸组和西药组大鼠血清IL-17、IL-23、IL-6、TNF-α、IL-1β均较模型组显著降低(P<0.05),而TGF-β均较模型组显著升高(P<0.05)。隔药组在下调大鼠血清IL-17、IL-23明显优于西药组(P<0.05),而在IL-6、TNF-α、IL-1β、TGF-β的调控上差异无统计学意义(P>0.05)。结论隔药灸能够调控UC大鼠血清Th17主要相关细胞因子的水平,这可能是隔药灸治疗UC的作用机制之一。Objective To explore the regulatory mechanism of herb-partitioned moxibustion of Thl7 major related cytokines on the serum of ulcerative colitis( UC) rats. Methods SD rats were randomly divided into normal group,model group,herb-partitioned moxibustion group and medicine group. A rat model of UC was made by an immunological method plus local stimulation. The serum levels of IL-17,IL-23,IL-6,TGF-β,IL-1β and TNF-α were detected by ELISA method. Result Serum levels of IL-17,IL-23,IL-6,IL-1β and TNF-α in UC rats were higher than those in the normal group( P〈 0. 05),whereas the serum level of TGF-β was lower than that of the normal group( P〈 0.05). After treatment,levels of IL-17,IL-23,IL-6,IL-1β and TNF-α of herb-partitioned moxibustion group and medicine group decreased obviously when comparing with model group( P〈 0. 05),and TGF-β increased obviously( P〈 0. 05) in the serum of both the herbpartitioned moxibustion group and medicine group comparing with the model group. Comparing with the medicine group,serum levels of IL-17 and IL-23 of the herb-partitioned moxibustion group decreased obviously( P〈 0. 05),and there was no statistical significance( P〈 0. 05)in the level of TGF-β,IL-1β and TNF-α. Conclusion Herb-partitioned moxibustion can regulate levels of Thl7 major related cytokines in the serum of UC Rats. This may be one of the mechanisms of herb-partitioned moxibustion treatment for UC.
