摘 要:目的 探讨脊髓核因子κB (NF-κB)抑制因子(IκB)/NF-κB信号转导通路在大鼠切口痛中的作用及机制.方法 将150只雄性清洁级Sprague-Dawley大鼠根据随机数字表法分为假手术组、二甲基亚砜(DMSO)组和脯氨酸二硫氨甲酸酯组(PDTC)组,各50只.鞘内穿刺成功后,假手术组鞘内注射0.9%氯化钠注射液0.25 ml;DMSO组鞘内注射20% DMS0 0.25 ml;PDTC组鞘内注射10 ng PDTC(PDTC溶解于0.25 ml 20% DMSO中).DMSO组和PDTC组制作切口痛模型.测定各组大鼠术前(To),术后4 h(T1)、8 h(T2)、16 h(T3)、24 h(T4)机械痛阈值(WMT);假手术组于相同时点测定WMT.WMT测定后,深麻醉处死大鼠,取大鼠腰段脊髓,采用酶联免疫吸附法检测大鼠脊髓组织NF-κB表达,逆转录聚合酶链反应检测大鼠IκBα mRNA、肿瘤坏死因子0(TNF-α)mRNA水平.结果 3组大鼠T0时点以及假手术组各时点WMT、NF-κB、IκBα mRNA、TNF-α mRNA水平差异均无统计学意义(均P>0.05).PDTC组和DMSO组T1 ~T4时点WMT均明显低于假手术组[(10.21±1.25)、(7.16±0.78)g比(15.25±2.14)g,(11.62±0.46)、(7.40±0.27)g比(15.53±1.84)g,(12.67±0.55)、(8.61±0.34)g比(15.82±2.25)g,(12.54±0.43)、(9.82 ±0.37)g比(15.26 ±2.38)g],NF-κB 、IκBα mRNA、TNF-α mRNA水平均明显高于假手术组[NF-κB:(3.31 ±0.43)、(5.13 ±0.86)比(1.02 ±0.26),(2.76±0.22)、(5.04±0.69)比(0.91±0.15),(2.25 ±0.34)、(4.29±0.62)比(1.06 ±0.17),(2.02±0.33)、(3.68±0.43)比(0.94 ±0.23);IκBαmRNA:(0.327±0.066)、(0.527±0.083)比(0.113±0.025),(0.302±0.034)、(0.503±0.072)比(0.127 ±0.021),(0.285 ±0.022)、(0.468±0.052)比(0.118 ±0.018),(0.257±0.030)、(0.453 ±0.046)比(0.125±0.014);TNF-α mRNA:(0.547±0.051)、(0.841±0.113)比(0.139±0.036),(0.502±0.046)、(0.783±0.094)比(0.138±0.02Objective To investigate the effect and mechanism of inhibitor κB (IκB)/nuclear factor κB (NF-κB) signal transduction pathway in spinal cord in rat incision pain mode.Methods One-hundred and fifty health adult male Sprague-Dawiey rats were randomly divided into sham operation group,dimethylsulfoxide (DMSO) group and pyrrolidine dithiocarbamate (PDTC) group (n =50 in each group).After successful intrathecal puncture,0.9% sodium chloride (0.25 ml),20% DMSO (0.25 ml) and PDTC (10 ng dissolved in 20% DMSO,0.25 ml) was respectively intrathecally injected in sham operation group,DMSO group and PDTC group.The model of incisional pain was established in DMSO group and PDTC group.The withdrawal mechanical threshold (WMT) of hindpaw was assessed before operation (T0),4 h (T1),8 h (T2),16 h (T3) and 24 h (T4) after surgery (at the same time pointed in sham operation group).After determination of WMT,the animals were executed under deep anesthesia to obtain the lumbar spinal cord.Enzyme linked immunosorbent assay was used to detect the expression of NF-κB in spinal cord tissue,and reverse transcriptase polymerase chain reaction was used to detect the mRNA levels of IκBα and tumor necrosis factor α (TNF α).Results The WMT,NF-κB expression,IκBα mRNA,TNF-α mRNA at T0 point were not significantly different among groups (P 〉 0.05).At T1-T4,WMT in PDTC and DMSO group were significantly lower than that in sham operation group [(10.21 ± 1.25),(7.16 ±0.78) g vs (15.25 ±2.14) g,(11.62 ±0.46),(7.40 ±0.27) g vs (15.53 ± 1.84) g,(12.67 ±0.55),(8.61 ±0.34) gvs (15.82±2.25) g,(12.54±0.43),(9.82 ±0.37) gvs (15.26± 2.38) g],the NF-κB expression,IκBα mRNA,TNF-α mRNA in PDTC and DMSO group were significantly higher than those in sham operation group [NF-κB expression:(3.31 ± 0.43),(5.13 ± 0.86) vs (1.02 ± 0.26),(2.76±0.22),(5.04±0.69) vs (0.91 ±0.15),(2.25±0.34)
