摘 要:目的:观察猴脑选择性超深低温断血流复苏对基质金属蛋白酶9(MMP-9)和转化生长因子β1(TGF-β1)表达的变化影响,探讨超深低温相关脑保护机制。方法:选择9只成年雄性健康恒河猴作为研究对象,随机分为常温组2只与超深低温组7只,分别建立等温及超深低温断血流复苏实验模型,分别对大脑皮质脑组织切片用MMP-9、TGF-β1抗体进行免疫组化染色、图像分析,对每张切片的平均灰度值进行统计学分析,比较两组MMP-9、TGF-β1含量和平均灰度值的情况。结果:超深低温组神经细胞MMP-9表达下调,而TGF-β1表达增高。结论:超深低温能抑制MMP-9的表达,增加TGF-β1的表达,促进受损脑细胞的损伤修复,提高了脑细胞对缺血缺氧损害的耐受性,抑制细胞的凋亡,是缺血缺氧情况下脑重要的内源性保护机制之一。Objective:To explore the effects of blood flow occlusion after resuscitation using selective cerebral ultra-profound hypothermia on the expression matrix metalloproteinases-9 and transforming growth factor-β1 for monkey and to investigate the mechanism of brain protection related to ultra deep hypothermia.Method:9 adult male healthy Ganges River monkeys were selected as the research subjects,randomly divided into the normal temperature group of two and the ultra deep low temperature group of seven,the isothermal and ultra deep hypothermic circulatory arrest recovery experimental model were established,and mmunohistochemical staining and image analysis were used to analyze the average gray value of each slice of brain cortex brain tissue sections with MMP-9 and TGF-β1 antibody,the MMP-9,TGF-β1 and the average gray value of the situation in two groups were compared. Result:The expression of MMP-9 was down regulated,while the expression of TGF-β1 was increased in the ultra deep hypothermia group.Conclusion:Selective ultra-profound hypothermia can attenuate MMP-9 expression and promote TGF-β1 expression which inhibit apoptosis,improve the tolerance of nerve cells to ischemia and hypoxia and promote the restoration of damaged nerve cells,it is one of the important endogenous protective mechanisms of cerebral ischemia and hypoxia.
