摘 要:目的:了解山茱萸对大鼠心肌缺血再灌注的保护作用机制。方法:选取84只雄性Wistar大鼠,按随机数字表法分三组,分别为对照组、缺血再灌注组(I/R组)和治疗组,每组28只。采用结扎大鼠冠状动脉左前降支法制备大鼠心肌缺血再灌注损伤模型,对照组在左冠状动脉前降支处仅穿线不结扎,I/R组在再灌注前5 min时尾静脉注射0.9%氯化钠注射液1 m L,治疗组在再灌注前5 min时尾静脉注射山茱萸药液1 m L,观察记录各组大鼠心肌缺血情况和心电图信息。结果:与对照组比较,I/R组及治疗组大鼠FS和EF均降低,LVDd、LVESV、LVEDV均上升;与I/R组比较,治疗组FS和EF均上升,LVDd、LVESV、LVEDV均下降,差异均有统计学意义(P<0.05)。结论:山茱萸及其有效成分通过PI3K/Akt通路,对大鼠心肌缺血再灌注损伤保护机制的实现起到了十分重要的作用。Objective:To learn the protective mechanism of action of cornus officinalis for rats with myocardial ischemia-reperfusion injury.Method:Eighty-four male Wistar rats were divided into three groups according to the random number table,the control group,the ischemia-reperfusion group(I/R group) and the treatment group,twenty-eight rats in each group.The model of myocardial ischemia-reperfusion injury in rats was developed by ligaturing the left descending anterior coronary artery,the rats in the control group were only thread while rats in the I/R group were given 1 m L 0.9% sodium chloride injection through the tail vein before reperfusion and rats in treatment group were given 1 m L cornus officinalis injection through the tail vein.The myocardial ischemia and electrocardiograph information of all groups were observated and recorded.Result:Compared with the control group,FS and EF of the rats in I/R group and treatment group were decreased,but LVDd,LVESV and LVEDV were increased,while compared with the I/R group,FS and EF of the rats in treatment group were increased,but LVDd,LVESV,LVEDV were decreased,the differences were all statistically significant(P〈0.05).Conclusion:Cornus officinalis may play a very important role for realizing the protective mechanism in rats with myocardial ischemia-reperfusion injury through PI3k/Akt pathway.
