香芹酚治疗COPD系统炎症大鼠模型的实验研究

摘  要:目的研究香芹酚对慢性阻塞性肺疾病(COPD)的预防作用。方法 30只雄性Wister大鼠随机分为六组,分别为:对照组、COPD组、地塞米松治疗组、低剂量香芹酚治疗组、中剂量香芹酚治疗组、高剂量香芹酚治疗组。除对照组外,大鼠暴露于香烟烟雾中3个月以制造COPD模型,A组大鼠饲养于干净环境中并且单独饮水;B组大鼠饲养于有香烟烟雾的环境中,并且单独饮水;C组大鼠暴露于香烟烟雾环境中并给予含地塞米松的水(50μg/ml);D组大鼠暴露于香烟烟雾环境中并且给予含低剂量香芹酚的水(60μg/ml);E组大鼠暴露于香烟烟雾环境中并且给予含中剂量香芹酚的水(120μg/ml);F组大鼠暴露于香烟烟雾环境中并且给予含高剂量香芹酚的水(240μg/ml)。检测各组血液中白细胞(WBC)的种类及血液中白细胞介素-8(IL-8)及丙二醛(MDA)水平。结果与对照组相比,COPD组血浆IL-8水平、MDA浓度、WBC、嗜酸性粒细胞均显著升高(P<0.01),而体重显著下降(P=0.004);与COPD组相比,地塞米松及中、高剂量香芹酚治疗组血浆IL-8水平、MDA浓度、WBC均显著下降(P<0.01),而体重显著提高(P<0.01);与地塞米松治疗组相比,中、高剂量香芹酚治疗组血浆MDA水平及IL-8浓度均显著下降(P<0.01)、体重均明显增高(P<0.001),WBC及嗜酸性粒细胞无明显变化。结论在大鼠COPD模型上,香芹酚可以预防病情进一步发展恶化,其作用与地塞米松的作用相当。Objective To investigate the role of carvacrol in preventing the inflammation of COPD. Methods Guinea pigs of both sexes were divided into 6 groups,including: control,COPD,COPD + drinking water containing three concentrations of carvacrol and COPD + dexamethasone group. In addition to the control group,the rats were exposed to cigarette smoke in the three months to manufacture COPD model,A group of rats were housed in a clean environment and water alone; group B rats were housed in cigarette fumes,and water alone; group C rats exposed to cigarette smoke in the environment and give water containing dexamethasone( 50 μg / ml); D group of rats exposed to cigarette smoke in the environment with low dose administered and carvacrol water( 60 μg / ml); E group of rats exposed to cigarette smoke in the environment and give a dose containing carvacrol water( 120 μg / ml); F group of rats exposed to cigarette smoke in the environment and to give high doses of carvacrol-containing water( 240μg /ml). White blood cell( WBC),interleukin-8( IL-8) and malondialdehyde( MDA) were measured. Results Serum levels of IL-8 and MDA,total WBC( P〈0. 01 for all cases) and eosinophil counts( P〈0. 05) increased and weight changes decreased( P〈0. 05) in COPD group compared to controls. Serum MDA level and total WBC in treated groups with two higher carvacrol concentrations,eosinophil,neutrophil and lymphocyte percentage in those treated with high concentration as well as IL-8 level and weight change in treated groups and in dexamethasone treated group were significantly improved compared to COPD group( P〈0. 001). Conclusion These results showed a preventive effect of the carvacrol on all measured parameter in COPD guinea pigs which was comparable to the effect of dexamethasone at used concentrations.
