摘 要:目的:建立去势后原发性膝骨关节炎大鼠模型,观察强骨宝对模型鼠关节软骨形态学及软骨基质的影响,为进一步研究提供基础。方法:1去势后原发性膝骨关节炎模型建立:麻醉下摘除双侧卵巢1周后行疲劳性跑步训练,跑台坡度-15°,频率28 m·min-1,60 min·d-1,每周6 d,建立膝骨关节炎模型。分别于2,4,6周后检测尿液中Ⅱ型胶原羧基端肽(CTX-Ⅱ),制作股骨髁病理切片鉴定模型。2动物分组与给药:将造模成功的大鼠随机分为模型对照组、强骨宝组、阳性对照组,分别给予相应处理。3取材与检测:给药4周后,取尿液、血清、双侧股骨髁,检测尿液中CTX-Ⅱ含量,股骨髁软骨经HE染色处理后行Mankin评分。结果:1模型鉴定:根据尿液CTX-Ⅱ含量及软骨Mankin评分结果,造模4周后关节软骨开始退变,6周后退变明显,模型成功。2给药治疗:Mankin评分及CTX-Ⅱ含量测定结果,与空白对照组比较,模型对照组、强骨宝组、阳性对照组均增高(P<0.01或P<0.05);与模型对照组比较,强骨宝组、阳性对照组均降低(P<0.05);与阳性对照组比较,强骨宝组差异无统计学意义(P>0.05)。结论:强骨宝可减缓原发性膝骨关节炎模型鼠的软骨损伤进程。Objective:To observe the effect of Qianggu Bao(强骨宝)on the articular cartilage of primary knee osteoarthritis in ovariectomized rats.Methods:(1)Establishing models of ovariectomized rats with primary knee osteoarthritis:After anesthesia,bilateral ovariectomy was performed for the experimental rats.After one week,they were forced to do fatigued running training on 15-degree treadmill with frequency of 28 m·min^-1,60 min·d^-1,for 6 days a week to establish knee osteoarthritis models.CTX-Ⅱ in urine was detected respectively after 2,4 and 6 weeks,making pathological identification model of femoral condyle.(2)Grouping and administration:Successfully modeled rats were randomly divided into a model group,a Qianggu Bao group and a positive control group,and each group was given corresponding treatment.(3)Sanjmpling and detection:After 4 weeks of administration,urine,serum and bilateral femoral condyle were sampled to detect the content of CTX-Ⅱ in urine and make Mankin scores to femoral condyle cartilage after HE staining.Results:(1)Model identification:According to the content of CTX-Ⅱ in urine and Mankin scores of femoral condyle cartilage,articular cartilage degeneration began after 4 weeks of modeling,which was obvious after 6 weeks,so the models were successfully established.(2)Administration for treatment:Compared with the blank control group,Mankin score and CTX-Ⅱ content of the model control group,the Qianggu Bao group and thepositive control group all increased(P〈0.01 or P〈0.05);compared with the model control group,those of the Qianggu Bao group and the positive control group decreased(P〈0.05);and the difference between the positive control group and the Qianggu Bao group had no statistical significance(P〉0.05).Conclusion:Qianggu Bao can slow down the process of cartilage damage in model rats with primary knee osteoarthritis.
