摘  要:为观察柴胡疏肝散对硝酸甘油偏头痛大鼠模型中脑导水管周围灰质(PAG)5-HT、NF-κB、C-fos表达的影响,以探讨其可能的作用机制。用60只SD大鼠随机分为6组:空白组、模型组、对照组及中药高、中、低剂量组,各组灌胃给予相应药液(或生理盐水),连续14 d,末次灌胃后以皮下注射硝酸甘油的方式复制偏头痛大鼠模型。通过蛋白印迹法观察偏头痛大鼠PAG NF-κB的表达;通过免疫组化法观察其5-HT、C-fos表达。结果预防性使用柴胡疏肝散能减少硝酸甘油偏头痛模型大鼠PAG NF-κB、C-fos的表达,提高硝酸甘油偏头痛模型大鼠PAG 5-HT的表达;临床等效剂量效果与氟桂利嗪相当(P>0.05),大剂量效果优于氟桂利嗪(P<0.05)。说明:1硝酸甘油偏头痛大鼠模型PAG NF-κB蛋白表达增加;2柴胡疏肝散能抑制硝酸甘油偏头痛大鼠NF-κB信号通路,减少其下游基因C-fos的表达,减轻神经源性炎症;该效应在一定剂量范围内具有量效关系;3硝酸甘油偏头痛大鼠模型PAG 5-HT表达降低;柴胡疏肝散能提高硝酸甘油偏头痛大鼠PAG 5-HT的表达,减少疼痛刺激伤害;该效应在一定剂量范围内具有量效关系。To observe the influence of the method of nourishing the liver and regulating blood and the formula on express of 5-HT,NF-κB and C-fos in PAG of cerebral aqueduct of the rat model of nitroglycerin type migraine,60 SD rats were divided into 6 group: blank group,model group,controlled group,high dose of decoction group,middose of decoction group and low-dose of decoction group. The decoction and normal saline was given to relevant group by gavage for 14 days. After the last gavage,the migraine rat model was copied by subcutaneous injection of nitroglycerin. To observe the express of NF-κB of PAG of migraine rat by the method of Western bolt,and to observe the express of 5-HT,C-fos by the method of immunohistochemistry. It decreases the express of NF-κB,C-fos in PAG of the nitroglycerin migraine rat model and increases the express of 5-HT in PAG of the rat model by using the decoction of Rougan Yupian preventively. It has the same effect by taking the same dose of this decoction and flunarizine( P 0. 05). It is better than flunarizine group by taking high-dose. Conclusion: 1 The express of NF-κB in PAG of the nitroglycerin migraine rat model is increased. The decoction of Rougan Yupian can inhibit the NF-κB code pathway of the nitroglycerin migraine rat model,decrease the express of C-fos which is downstream target gene and relieve the neurogenic inflammation. The effect has some dose-effect relationship in some dose range. 2 The express of 5-HT in PAG of the nitroglycerin migraine rat model is decreased. The decoction of Rou Gan Yu Pian can improve the express of 5-HT in PAG of the nitroglycerin migraine rat model and decrease the hurt of pain. The effect has some dose-effect relationship in some dose range.
