丝瓜提取物对衰老小鼠学习记忆能力、脑组织形态学及免疫功能的影响

摘　　要：目的:观察丝瓜提取物对衰老小鼠学习记忆能力、脑组织形态学及免疫功能的影响。方法:经筛选的昆明种小鼠随机分为对照组、衰老组、丝瓜提取物187.5 mg/kg、375 mg/kg、750 mg/kg 3个剂量组。除对照组以外,均在颈背部皮下持续注射D-半乳糖复制衰老模型,利用跳台实验及Morris水迷宫测试各组小鼠学习记忆能力,脏体比方法测定各组小鼠脑指数、胸腺指数和脾脏指数,ELISA法检测各组小鼠血清中细胞因子IL-2,IFN-r的浓度和T淋巴细胞增殖活性,HE染色法观察各组小鼠脑组织形态学变化。结果:衰老组在跳台实验的能力测试中错误次数增多;在水迷宫测试中逃避潜伏期延长,平台象限游泳时间缩短,穿越平台次数减少;脑组织细胞数量明显减少;脑指数、胸腺指数和脾脏指数减少;IL-2、IFN-r水平和T淋巴细胞增殖活性降低;与衰老组比较,丝瓜提取物(375 mg/kg、750mg/kg)可明显拮抗上述指标变化。结论:丝瓜提取物可以改善衰老小鼠的学习记忆,同时增加机体免疫活性,达到延缓衰老进程的作用。Objective： To observe the towel gourd extract on aging mice learning memory ability,the influence of the brain morphology and immune function. Methods： After the screening of kunming mice were randomly divided into control group,aging group,towel gourd extract 187. 5mg / kg dose groups,375 mg / kg dose groupsand 750 mg / kg dose groups,aging model were induced by injection of D-galactose inthe back of the neck. The ability of learning and memory were evaluated by the platform experiment and Morris water maze test. Brain index,thymus index and spleen index were determinedby the proportion of viscera and body weight. The type cytokines in serum IL-2,the concentration of IFN-r and the activity of T lymphocyte proliferation were measurementedby ELISA. Brain tissue morphology in mice were observedby HE staining.Results： Aging group increased the number of errors in the experiment jumping ability test. in the Morris water maze test escape latency was prolonged,the platform quadrant swimming time is shortened,reduced frequency of crossing platform. significantly reduced the number of brain cells. reduced brain index,thymus index and spleen index. IL-2,IFN-r level and T lymphocyte proliferation activity is decreased. Comparedwith the aging group,towel gourd extract（ 375 mg / kg,750 mg / kg） can antagonize the changes of above indexes. Conclusion： Towel gourd extract can improve the learning and memory of aging mice,increase the body＇s immune activity at the same time and slow down the ageing process.
