摘  要:目的:研究地黄益智颗粒(简称地黄益智)对老年性痴呆的防治作用,并对其作用机理进行初步探讨。方法:腹腔注射D-半乳糖120 mg/kg每天1次,连续8周诱导大鼠痴呆模型,与此同时分别灌胃地黄益智颗粒5、10、20g/kg,吡拉西坦0.7 g/kg,对照组给予等量蒸馏水,每天1次,连续8周。在给药后4周、8周时用Morris水迷宫测定其学习记忆能力,并于试验结束时测定其血液以及脑组织的生化指标。结果:连续注射D-半乳糖4周后,模型组大鼠在定向航行试验其寻找到平台的潜伏期以及持续时间在第一天和第三天时显著延长,在空间探索试验中进入原平台区域的潜伏期明显延长,与对照组比较具有统计学意义。地黄益智5、10、20g/kg能明显的缩短D-半乳糖痴呆大鼠在定向航行试验中第二、三天的潜伏期时间,显著缩短空间探索的潜伏期,与模型组比较具有统计学差异。连续注射D-半乳糖8周,可显著延长大鼠在定向航行试验中寻找到平台的潜伏期暨持续时间,显著延长大鼠在空间探索试验中找到原平台区域的潜伏期以及进入该区域的次数,其血液SOD水平降低、MDA水平升高,脑组织Ach E水平及NO水平升高、Ch AT活性降低,与对照组比较差异具有统计学意义;地黄益智5、10、20g/kg和吡拉西坦0.7 g/kg可显著缩短D-半乳糖大鼠在定向航行试验第一、二天的持续时间以及潜伏期,并显著改善其血液及脑组织上述生化指标。结论:地黄益智对老年性痴呆有一定的防治作用,其机理可能与其可以降低氧化应激水平和增加神经突触间的乙酰胆碱浓度有关。Objective: To evaluate the preventive effect of Dihuang Yizhi Granules( referred Dihuang Yizhi) on Alzheimer's disease( AD),and explore its mechanism of action. Mthods: Rats were induced dementia by intraperitoneal injection of D-galactose 120 mg / kg,once per day for 8 weeks induced rat models of dementia,meanwhile the rats were intragastric administration of Dihuang Yizhi Granules 5,10,20 g / kg,piracetam 0. 7 g / kg or the same volume of distilled water. 4 weeks and 8 weeks later,the learning and memory abilities of rats were checked with Morris water maze,and the biochemical indices were also measured in the blood and brain tissue in rats at the end of the test. Results:Intraperitoneal injection of D-galactose 120 mg / kg for four weeks significantly prolonged the latent period and persistent period in place navigation test during the first day and third day and the time of enter into the original platform in the space exploration test in rats. Dihuang Yizhi5,10,20 g / kg can significantly shorten the latent period and persistent period during the second day and third day in place navigation test and the time of enter into the original platform in the space exploration test in D-galactose rats. 8 weeks later,the latent period and persistent period in place navigation test or in the space exploration test in the D-galactose rats were much longer than the control group,moreover the levels of SOD was reduced but MDA was increased in blood,the levels of Ach E and NO were increased,the activity of Ch AT was decreased in brain in the D-galactose rats. Dihuang Yizhi 5,10,20 g / kg can significantly shorten the latent period and persistent period in place navigation test during the first day and second day,and significantly improve the biochemical indicators in the D-galactose rats. Conclusion: Dihuang Yizhi has therapeutic effect on AD,it may be relate to protect the central nervous tissue from oxidative stress.
