摘  要:目的繁殖和鉴定ApoE^(-/-)小鼠,建立血管损伤模型,为进一步研究动脉粥样硬化奠定基础。方法将引进的小鼠采用1只雌性与1只雄性同笼的方式进行繁殖。从子代小鼠尾巴中提取DNA,用PCR法和琼脂糖凝胶电泳法鉴定小鼠的基因型。获取ApoE^(-/-)纯合子小鼠,并绘制其0~20周龄生长曲线。采用油红O脂类染色法从血管形态学方面观察ApoE^(-/-)小鼠。结果成功繁育出的ApoE^(-/-)纯合小鼠较C57小鼠生长缓慢,且20周龄ApoE^(-/-)小鼠血管有明显损伤。结论利用ApoE^(-/-)纯合子小鼠能够建立良好的血管损伤模型。To breed and identify apolipoprotein E( Apo E) gene knock out mice for further study of atherosclerosis with dyslipidemia model,apolipoprotein E- /-mice were bred and reproduced. Genomic DNA was isolated from tails and subjected to PCR and agarose gel electrophoresis for offspring genotype identification. The growth rate of ApoE-/-homozygous mice during 0—20 weeks of age were determined. The vascular morphological of ApoE-/-mice were observed by Oil Red O dye method. The results showed that ApoE-/-mice was successfully bred. The growth of ApoE-/-mice was slowly than wild type C57 BL /6 mice. And the obvious damage of vascular in ApoE-/-mice at 20- week age was obvious. Conclusion The damaged vascular model of ApoE-/-mice was successfully established.
