摘 要:目的:优选聚酰胺树脂分离纯化"前愈"总黄酮的工艺条件。方法:采用紫外分光光度法,以总黄酮含量为指标,利用静态和动态结合的方法考察上样质量浓度、pH、树脂药材比、洗脱溶剂的选择及用量等工艺参数,评价聚酰胺树脂纯化总黄酮的行为,确定其最佳纯化工艺。结果:最佳纯化工艺参数为上样液质量浓度1.0 g·m L^(-1),pH 3.0,5 BV水洗脱除杂,洗脱剂50%乙醇,洗脱剂用量4 BV;此工艺条件下,总黄酮纯度由8.89%提高到63.67%,转移率25.51%。结论:采用聚酰胺树脂纯化"前愈"中总黄酮的效果良好,为后期复方中总黄酮部位的活性研究及新剂型的开发提供基础。Objective: To optimize technology for purification of total flavonoids from compound"Qianyu"with polyamide resin. Methods: The purity of total flavonoids was determined with the UV spectrophotometer as an index. Purification behavior of polyamide resin for total flavonoids was investigated under static and dynamic conditions. The best purification technology was established by investigating the concentration of sample solution,p H,ratio of resin and herbs,elution volume and other factors. Results: The best purification technology of total flavonoids involved 1. 0 g·m L^(- 1)crude drug solution,p H 3. 0,5 BV water for removing the impurities,and 4 BV50% ethanol for eluting total flavonoids. Under these conditions,purity of total flavonoids was up to 63. 67% from8. 89%,and the transfer rate was 25. 51%. Conclusion: Purification of total flavonoids from compound "Qianyu"with polyamide resin is feasible; this could provide a basis for activity investigation of total flavonoids from compound "Qianyu"and develop new dosage forms.
