摘  要:目的:研究黄花柳花总黄酮对在体心肌缺血再灌注损伤大鼠模型是否具有保护作用并探讨其相关机制。方法:结扎大鼠心脏左冠状动脉前将支30 min,造成缺血状态后再灌2 h,实时监测心电图ST段变化。采用腹主动脉采血,检测血清中LDH,CK-MB,AST,GSH-PX,SOD,MDA,MPO,IL-6和TNF-α等相关指标变化,利用苏木素-伊红(HE)染色观察心肌组织病理学改变。结果:与模型组比较,给药组大鼠血清中心肌酶LDH,CK-MB和AST含量降低,氧化指标GSH-Px,SOD含量升高,MDA含量降低,炎症因子MPO,IL-6和TNF-α降低,病理特征有明显改善。结论:维药黄花柳花总黄酮对心肌缺血再灌注损伤具有一定的保护作用。Objective: To evaluate the protective effect of total flavones from Salix caprea flowers( TFSC)on myocardial ischemia-reperfusion injury( MI / RI) in rats,and explore the underlying mechanisms. Methods:Rat hearts were exposed and the left anterior descending coronary artery( LAD) was ligated for 30 min followed by2-h reperfusion. The levels of lactate dehydrogenase( LDH),creatine kinase isoenzyme( CK-MB),superoxide dismutase( SOD),and malondialdehyde( MDA),acetic transaminase( AST),total superoxide dismutase( TSOD),glutathi-one peroxidase( GSH-Px),myeloperoxidase( MPO),malonaldehyde( MDA),nterleukins( IL-6),tumor necrosis factor-α( TNF-α) in serum were detected. S-T segment change was recorded and the histopathological changes were observed. Results: Compared with MI / RI control,pretreatment with TFSC in vivo significantly improved the hemodynamics,decreased MDA,increased the activities of plasma SOD and GSH-Px,and attenuated S-T segment elevation and myocardium pathological changes. It also decreased the activities of LDH,CK,CK-MB,AST and MPO,and the levels of IL-6 and TNF-α in serum. Conclusion: TFSC has benefits to protect against the heart injury induced by ischaemia / reperfusion.
