摘 要:目的观察盐酸吗啡对人乳腺癌MCF-7细胞生长增殖的影响。方法人乳腺癌MCF-7细胞培养至对数生长期,采用随机数字表法,将其随机分为4组:空白对照组(C组)、0.01μmol/L吗啡组(M1组)、0.10μmol/L吗啡组(M2组)、1.00μmol/L吗啡组(M3组)。M1、M2、M3组分别在培养液中加入吗啡,使吗啡的终质量浓度分别为0.01、0.10、1.00μmol/L;对照组(C组)不加入任何药物。噻唑蓝(MTT)法和克隆形成实验观察细胞生长增殖的变化,流式细胞仪检测细胞周期和细胞凋亡。结果与C组比较,M1、M2、M3组人乳腺癌MCF-7细胞的生长速度减慢,其克隆形成率分别为(0.76±0.09)%、(0.74±0.12)%和(0.78±0.08)%,低于C组的(1.25±0.28)%(P〈0.05),细胞凋亡率分别为(5.33±0.09)%、(5.21±0.15)%和(5.09±0.08)%,较C组的(1.25±0.16)%升高(P〈0.05),而且细胞周期G2/M期的比例增加,S期比例减少(P〈0.05);M1组、M2组、M3组两两比较差异无统计学意义(P〉0.05)。结论盐酸吗啡促进人乳腺癌MCF-7细胞的凋亡,使细胞周期停滞于G2/M期,从而抑制乳腺癌细胞的生长增殖,且其抑制作用有封顶效应。Objective To observe the effects of morphine on the growth and proliferation of human breast cancer MCF - 7 cells. Methods MCF - 7 cells were seeded in 6 - well or 96 - well plates and divided into 4 groups : 0. 01 izmol/L morphine group ( group M1 ), 0. 10 μmol/L morphine group ( group M2 ), 1.00 μmol/L morphine group ( group M3 ) and control group ( group C ). MCF - 7 cells in the logarithmic phase were cultured. The cells in morphine groups were incubated with 0. 01, 0. 10 or 1. 00 μmol/L mor- phine respectively. The cells in control group were incubated normally. The viability of cells was deter- mined by methyl thiazol tetrazolium (MTF) assay and the proliferation of cells was determined by colony formation assay. The cell cycle progression and apoptosis were assessed by flow eytometry. Results The viability of MCF - 7 cells was inhibited after exposed to 0. 01, 0. 10 or 1.00 μmol/L morphine respective- ly. Cell colony formation rate in groups M1, M2, and M3 was (0. 76 ±0.09)% , (0.74 ±0. 12)% and (0. 78 ± 0. 08 ) %, respectively, which significantly lower than in group C [ ( 1.25 ± 0. 28 ) % ] ( P 〈 0.05). Theapoptotic rate in groups M1, M2, and M3 was (5.33 ±0.09)%, (5.21 ±0.15)% and (5.09 ±0. 08) %, respectively, significantly higher than that in control group [ ( 1.25 ±0. 16) % ] ( P 〈 0. 05 ). In morphine groups, the proportion of cells in G2/M was significantly higher than that in group C (P 〈 0.05 ). There was no significant difference in the above indexes among the three morphine groups ( P 〉 0. 05). Conclusion Morphine can inhibit growth and proliferation of MCF - 7 cells by inducing ap- optosis and making cell cycle arrest in vitro. But morphine presents a ceiling effect on inhibiting growth and proliferation of breast cancer cells MCF -7.
