M2型巨噬细胞向M1型巨噬细胞的转化及其意义

摘　　要：目的诱导人单核细胞向M2型巨噬细胞转化及探讨诱导M2型巨噬细胞向M1型巨噬细胞转化的可行性。方法通过磁珠阳性分选法从健康成人新鲜血液中获得人单核细胞，经重组人巨噬细胞集落刺激因子（M-CSF）诱导6d，应用流式细胞仪鉴定M2型巨噬细胞标志物CD163阳性表达率。对M2型巨噬细胞应用干扰素（IFN）-γ和脂多糖（LPS）诱导2～3d，应用流式细胞仪鉴定M1型巨噬细胞表面标志物CD16阳性表达率。结果经重组人M-CSF诱导后的单核细胞，M2型巨噬细胞的CD163阳性表达率为（66．37±2．67）％，MI型巨噬细胞的CD16阳性表达率为（13．37±4．41）％，M2型巨噬细胞阳性表达率明显高于M1型巨噬细胞阳性表达率（P〈0．01）。而M2型巨噬细胞再经IFN-γ和LPS诱导后，M1型巨噬细胞的CD16阳性表达率为（48．57±5．98）％，M2型巨噬细胞的CD163阳性表达率为（25．83±1．95）％，M1型巨噬细胞的阳性表达率明显高于M2型巨噬细胞的阳性表达率（P〈0．05）。结论人M2型巨噬细胞在IFN-γ和LPS诱导下可向M1型巨噬细胞转化。Objective To induce the human mononuclear cells to type M2 macrophages and to in- vestigate the feasibility of inducting type M2 macrophages to M1 macrophages. Methods Human mononu- clear cells were obtained through the positive magnetic bead separation method from fresh blood, and then induced for 6 days by human recombinant macrophage - colony stimulating factor （ M - CSF）. Flow cytom- etry was used to identify the M2 macrophages which were marked with CD163. M2 macrophages were in- duced for 2 - 3 days by interferon - γ （ IFN -γ） and lipopolysaccharide （LPS）. Flow cytometry was used to identify the M1 macrophages which were marked with CD16. Results Human mononuclear cells were induced by human recombinant M - CSF. The CD163 positive expression rate of M2 maerophages was （66. 37 ± 2. 67 ） % , and the CD16 positive expression rate of M1 macrophages was （ 13. 37 ± 4. 41 ） % （P 〈0. 01 ）. After induction by IFN - γ and LPS, the CD16 positive expression rate of M1 macrophages was （48.57 ± 5.98）%, and the CD163 positive expression rate of M2 macrophages was （25.83 ± 1.95 ） % （P 〈 0. 05 ）. Conclusion M2 macrophages could be transformed to M1 macrophages by IFN - γ and LPS.
