摘  要:目的:探索"体外去细胞-明胶再基质化-碳化二亚胺/N-羟基琥珀酰亚胺(以下均简称EDC/NHS)交联-体内自体细胞植入"的"嵌合型组织重构理论"在牛心包片处理过程中的科学性和可行性,以寻找组织工程生物瓣膜理想的支架材料。方法:采用"明胶再基质化-EDC/NHS交联"对脱细胞牛心包组织进行嵌合处理后植入大鼠皮下,通过HE染色对比新鲜组、脱细胞组、戊二醛组、嵌合组植入前后组织形态学改变,并对各组牛心包片进行厚度检测、含水量和力学检测、钙含量测定试验,判断该嵌合方法对牛心包基质的影响。结果:基线水平抗张强度测定结果显示,嵌合组牛心包抗张强度与戊二醛组(P=0.09)及脱细胞组(P=0.56)相比较并无显著差异远低于新鲜组(P=0.001);钙含量远低于新鲜组(P<0.001)及戊二醛组(P<0.001)。包埋2月后,嵌合组牛心包钙含量远低于新鲜组(P<0.001)及戊二醛组(P<0.001)。大鼠机体对明胶嵌合组处理牛心包的浸润程度最轻,然后是戊二醛组、脱细胞组,而新鲜组的浸润程度最重。在大鼠皮下降解率明显低于脱细胞组,炎症细胞浸润程度显著低于新鲜组,与戊醛组相当。结论:作为瓣膜组织工程支架材料,嵌合型牛心包膜比戊二醛交联效果具有更好生物相容性,钙化降解低,是较好的交联方法。Objective: We aimed to validate chimeric tissue remodeling on the tissue engineering production of bovine pericardium,but also to Looking for an ideal biological valve tissue engineering scaffolds.Methods: Acellular bovine pericardium tissue were implanted subcutaneously in rats after " re-gelatin matrix of EDC / NHS crosslinking" process,the morphological changes,thickness,moisture and mechanical testing and calcium content were analyzed among fresh group,decellularized group,glutaraldehyde group and matrix group.Results: Tensile strength measurement results showed chimeric group is far lower than the fresh group( P =0. 001),but when compared with glutaraldehyde group( P = 0. 09) and decellular group( P = 0. 56),there were no significant difference. The water content of the chimeric group was much higher than the fresh group( P= 0. 05),but there were no significant differences when compared with glutaraldehyde group( P = 0. 06) and decellular group( P = 0. 79). The calcium contentwas far lower than the fresh group( P〈0. 001) and glutaraldehyde group( P〈0. 001). After embedding in 60 days,the calcium content of the chimeric bovine pericardium group is far lower than the fresh group( P〈0. 001) and glutaraldehyde group( P〈0. 001). Chimeric bovine pericardium group shown the lightest Infiltration,then glutaraldehyde group,decellularized group,but the fresh group of the heaviest degree of infiltration. Degradation was significantly lower than acellular group,inflammatory cell infiltration was significantly lower than fresh group,but comparable with glutaraldehyde group.Conclusion: As the valve tissue engineering scaffolds,chimeric bovine pericardium over glutaraldehyde with better biocompatibility and lower calcification degradation,expressed a better crosslinking process.
