miRNA-146a在颈动脉粥样硬化患者血浆的表达及意义

摘　　要：目的研究颈动脉粥样硬化患者血浆中小分子RNA-146a(miR-146a)与斑块稳定性之间的关系。方法选择可能存在颈动脉硬化的患者,根据颈动脉超声检测结果分为正常组、稳定斑块组和不稳定斑块组,常规检测血压、体质量指数、肝肾功能、血糖、血脂、尿酸、超敏C反应蛋白(hsCRP),采用ELISA法检测血浆中IL-1水平,采用real time PCR检测血浆中的miRNA-146a的表达水平。结果 3组患者的年龄、体质量指数、血压、血糖、血脂、尿酸、肝肾功能差异均无统计学意义(P>0.05);与正常组比较,不稳定斑块组患者的血清hsCRP及血浆IL-1均明显升高(P<0.05);稳定斑块组和不稳定斑块组患者血浆中的miR-146a水平均明显升高(P<0.05),不稳定斑块组升高更加显著(P<0.01)。miR-146a与hsCRP呈弱正相关,与IL-1呈强正相关。结论 miR-146a可能参与了颈动脉粥样硬化发生过程。Objective To investigate the association between miRNA-146 a and carotid atherosclerosis. Methods106 cases in our hospital were divided into normal group,stable plaque group and unstable plaque group according to carotid ultrasonography. The levels of blood pressure,body mass index（ BMI）,liver function（ ALT）,renal function（ serum creatinine）,fasting blood glucose,blood fat,uric acid and high sensitive C-reactive protein（ hsCRP） were measured.ELISA was used to measure the plasma level of interleukin-1（ IL-1）.The plasma level of miRNA-146 a was detected by real time polymerase chain reaction technology. Results There were no significant differences in age,blood pressure,BMI,ALT,serum creatinine,blood glucose,blood fat between three groups（ P0. 05）. Compared with normal group,the levels of hsCRP and IL-1 in unstable plaque group significantly increased（ P 0. 05）. The plasma level of miRNA-146 a in the patients with plaque was significantly increased,compared with normal group（ P 0. 05）,especially in the patients with unstable plaque（ P0. 01）. There was mild positive association between plasma miRNA-146 a and serum hsCRP,and there was strong positive association between plasma miRNA-146 a and plasma IL-1. Conclusions miRNA-146 a may be related with the pathological progress of carotid atherosclerosis.
