减味寿胎丸及其君药菟丝子提取物对人早孕滋养层细胞影响的实验研究

摘　　要：目的探讨减味寿胎丸提取物、菟丝子总提物的含药血清对正常人早孕细胞滋养层细胞(CTB)增殖及凋亡的调节作用。方法制备减味寿胎丸提取物、菟丝子总提物的含药血清,研究其对CTB细胞增殖、分化潜能及凋亡的影响。结果除5%菟丝子总提物高剂量组外,不同组别不同浓度的含药血清与空白对照组血清相比,吸光度值均有显著统计学差异(P<0.05或P<0.01)。菟丝子总提物高、低剂量组5%与20%含药血清吸光度值有显著统计学差异(P<0.05)。各提取物10%含药血清分别培养细胞24 h后,S期细胞百分比均较空白对照组显著增加,G2/M期细胞百分比显著减少(P<0.05或P<0.01);干预48 h后,变化更加明显。结论菟丝子总提物高、低浓度的含药血清均可增加细胞的增殖活性,且呈明显的剂量依赖性。10%含药血清培养细胞24、48 h后,处于S期的细胞明显增多,阻滞在G2-M期细胞减少,表明含药血清可诱导细胞处于DNA合成期及有丝分裂期,促进细胞增殖,降低细胞凋亡的发生率。Objective To observe the effects of serum of the component of Modified Shoutai Wan（ MSTW） recipe and its principal drug semen cuscutae on the cell proliferation,differentiation and apoptosis of the human cytotrophoblast（ CTB） cell model. Methods Prepare the drug contain serum of the MSTW recipe and semen cuscutae,and explore their effects on the proliferation potential and apoptosis of the CTB cells. Results In addition to the 5% of total dodder high-dose group,different groups of different concentrations of medicated serum compared with blank control group serum,were significantly different from that of the blank control group（ P〈0. 05 or P〈0. 01）. Dodder high and low dosage group of total 5% and 20% medicated serum absorbance values have a statistically significant difference（ P〈0. 05）.After treated with 10% serum of different extracts for 24 hs,the percentage of S-hase were obviously accelerated than the blank group,and the percentage of G2 / M phase were obviously reduced than the blank group（ P〈0. 05 or P〈0. 01）.Changes of 48 hs treatment were accordance with the 24 hs. Conclusion High and low concentrations of serum of the extracts can increase the proliferative activity of the CTB cells in an obvious dosage dependent tendency. After treated with10% serum of different extracts for 24hs-and 48 hs respectively,the percentage of S-phase were obviously accelerated and that of G2 / M phase were obviously reduced,which indicated that serum of the extracts can induce cells into the DNA synthesis phase and mitosis phase,so as to promote the proliferation activity and reduce the incidence apoptosis.
