槲皮素调控内质网应激抑制H_2O_2诱导脐静脉内皮细胞凋亡的研究

摘  要:目的研究槲皮素对过氧化氢(H2O2)诱导脐静脉内皮细胞(HUVECs)凋亡的影响。方法体外培养脐静脉内皮细胞,H2O2作用于HUVECs诱导其凋亡,采用MTT法检测细胞活力,流式细胞术检测细胞凋亡率,Western-blot检测GRP78、XBP1及caspase-3蛋白表达。结果 H2O2呈剂量依赖性降低内皮细胞活力,并引起细胞凋亡。其中500μmol/L H2O2作用于细胞后,与正常组相比,细胞活力明显受到抑制,且GRP78、XBP1、caspase-3等蛋白含量明显增加(P<0.01);槲皮素预处理后,随着槲皮素浓度增加,细胞活力随之增加;与模型组相比,10μmol/L槲皮素能够显著抑制细胞凋亡,GRP78、XBP1、caspase-3等表达降低(P<0.01)。结论槲皮素能抑制H2O2诱导的内皮细胞凋亡,该作用可能与下调GRP78、XBP1、caspase-3等表达有关。Objective To study the effect of quercetin on the apoptosis of human umbilical vein endothelial cells in vitro induced by hydrogen peroxide( H2O2). Methods Human umbilical vein endothelial cells were cultured in vitro,and the apoptosis model was established by hydrogen peroxide. MTT was used to detect the growth activity of the cells,and flow cytometry was used to detect the apoptosis rate,GRP78,XBP1 and caspase-3 protein were detected by Western-blot. Results Hydrogen peroxide could decrease the activity of endothelial cells in a dose-dependent manner,and induce apoptosis. Among them,after 500 μmol / L H2O2,cell proliferation was obviously inhibited,and the content of GRP78,XBP1 and caspase-3 were significantly increased( P〈0. 01). After the addition of quercetin,the survival rate of cells increased with the increasing of the concentration of quercetin,and compared with the H2O2 model,10 μmol / L of quercetin pretreatment could significantly inhibit apoptosis,and decrease the expression of GRP78,XBP1 and caspase-3( P〈0. 01). Conclusion Quercetin can inhibit the apoptosis of endothelial cells induced by hydrogen peroxide,which may be related to the down-regulation of GRP78,XBP1 and caspase-3.
