摘 要:目的探讨氟中毒大鼠肝脏组织中Caspase-3及Cleaved-Caspase-3的表达情况。方法选用36只健康SD大鼠分为对照组、低氟组、高氟组(饮用含氟量分别为<1mg/L、5mg/L及50mg/L的自来水),每组12只(雌雄各半)。饲养6个月后,股动脉放血处死,氟离子电极法检测大鼠尿氟含量;自动血生化分析仪检测大鼠肝功能;免疫组织化学法和蛋白印记(Western blot)法检测Caspase-3及Cleaved-Caspase-3蛋白表达水平。结果低氟组、高氟组大鼠尿氟[(1.85±0.13)、(2.23±0.10)mg/L均高于对照组[(1.63±0.11)mg/L](P<0.05);低、高氟组大鼠血清谷丙转氨酶及谷草转氨酶活性[(48.66±5.55)、(68.17±8.39)U/L及(142.57±16.21)、(165.89±28.26)U/L]均高于对照组[(38.49±2.98)、(117.98±9.88)U/L](P<0.05);高过剂量氟组Caspase-3及Cleaved-Caspase-3蛋白表达[(98.08±13.08)、(106.33±9.14)]明显高于对照组[(53.92±6.36)、(59.78±7.54)](P<0.05)。结论氟可促进Caspase-3及Cleaved-Caspase-3蛋白的表达,其可能参与慢性氟中毒所致肝脏损伤发病过程。Objective To explore the role of Caspase-3and Cleaved-Caspase-3in rats liver damaged by excessive fluorine.Methods Thirty-six healthy Sprague-Dawley(SD)rats were randomly divided into three groups which were control group(drunk water containing NaF1mg/L),low and high fluorosis groups(drunk water containing NaF of 5mg/L and 50mg/L).After 6months of experiment treatment,the fluorine contents of urine were detected by fluorin-ion electrode method.The rats liver function determined by automatic blood chemical analyzer.The protein expression of Caspase-3and Cleaved-Caspase-3were detected by immunohistochemistry(IHC)and protein imprinting(western blot).Results The fluorine contents of urine in low and high-dose fluoride groups [(1.85±0.13),(2.23±0.10)mg/L]was higher than control group [(1.63±0.11)mg/L](P〈0.05).The activity of serum ALT and AST in low and high-dose fluorosis group[(48.66±5.55),(68.17±8.39)U/L and(142.57±16.21),(165.89±28.26)U/L]was higher than control group[(38.49±2.98),(117.98±9.88)U/L](P〈0.05).The protein expression of Caspase-3and Cleaved-Caspase-3 [(98.08±13.08),(106.3±9.14)]in high-dose fluorosis group were significantly increased than control group [(53.92±6.36),(59.78±7.54)](P〈0.05).Conclusion Caspase-3and Cleaved-Caspase-3may become the pathogenesis of liver damage in chronic fluorosis.
