摘  要:采用响应面法对海洋微生物Pseudozymasp.SD301发酵产角鲨烯培养条件进行优化。在单因素实验结果的基础上,通过Design-Expert软件对葡萄糖浓度、接种量、培养温度、装液量进行4因素响应面实验设计,以角鲨烯产量为响应值,优化菌株Pseudozymasp.SD301的培养条件,并验证响应面预测值与实测值的一致性。结果表明,在葡萄糖浓度为67.2g·L-1、接种量为1.5%、培养温度为26℃、装液量在41mL/250mL的最优培养条件下,角鲨烯产量为1.152g·L-1,比出发条件提高了3.62倍,实测值与响应面预测值吻合良好。Fermentation conditions for squalene production by marine microbe Pseudozymasp.SD301 were optimized in shake flask using response surface methodology(RSM).Based on the results obtained from the single factor tests,response surface analysis was designed with Design-Expert.The effects of glucose concentration,inoculation quantity,culture temperature and loading quantity on the yield of squalene were studied using RSM.By the central composite design,apredictive polynomial quadratic model was set up and the optimum fermentation conditions were developed.The theoretical optimal culture conditions are as follows:concentration of glucose was 67.2g·L^-1,inoculation quantity was 1.5%(v/v),culture temperature was 26 ℃,loading quantity was 41 mL in a 250 mL flask.Under these conditions,the yield of squalene reached 1.152g·L^-1,which is consistent with the predicted value of1.1508g·L^-1,representing a 3.62-fold increase compared to the original conditions.
