摘  要:为研究阿莫西林(AMO)在鸡排泄物中的残留消除规律,将鸡排泄物先经乙腈提取、饱和二氯甲烷萃取,上清液在酸性条件下经水杨醛沸水浴衍生化后,于激发波长354nm、发射波长445nm处用高效液相色谱荧光检测器检测。结果表明:该方法测定鸡排泄物中AMO的检测限为3.0ng·g^(-1)(S/N=3)、定量限为6.0ng·g^(-1)(S/N=10)。AMO在鸡排泄物样品中平均回收率为71.52%~81.23%,相对标准偏差均低于8.61%。将京海黄鸡分成2组,以25.0、50.0mg·(kg·d)^(-1)剂量内服AMO,每天给药1次,连续5d。投药第1天AMO在鸡排泄物中残留量即可检测到,投药第5天达峰值。休药后AMO在鸡排泄物中残留量迅速降低。低剂量组,休药第5天鸡排泄物中AMO残留量低于检测限;高剂量组,休药第7天鸡排泄物中AMO残留量低于检测限。鸡排泄物中AMO残留量与给药剂量呈正相关,鸡排泄物中高含量AMO残留存在较大的环境风险。A study on amoxicillin(AMO)residue depletion was conducted in chicken excreta.AMO was deproteinized from chicken excrements with acetonitrile,the analyte was extracted by saturated dichloromethane.The supernatant was reacted with salicylaldehyde under acidic and boiling conditions.Finally,AMO was determinated by high performance liquid chromatography(HPLC)with fluorescence detector.The excitation wavelength was set at 354 nm and emission wavelength was set at 445 nm.The average recovery in chicken excreta was 71.52%-81.23% with coefficients of variation(CV)lower than 8.61%.The limit of detection(LOD)was 3.0ng·g^-1(S/N=3)and the limit of quantitation(LOQ)was 6.0ng·g^-1(S/N =10).After the chickens were orally administered successively AMO of 25.0and50.0mg·kg^-1 of body weight one time every day for 5days,AMO residues in chicken excreta were detected at the first day,the maximum residues of AMO in chicken excreta were detected in the 5th day.The low-dose group was lower than LOD(6.0μg·kg^-1)at the 5th withdrawal day and the high-dose group was lower than LOD at the 7th withdrawal day.The residues of AMO in chicken excreta were all positively correlated with AMO orally administered doses.There are environmental risks with high residues of AMO in chicken excreta.
