摘  要:目的探索大鼠骨髓间充质干细胞诱导分化为神经样细胞的新方法,为进一步动物实验研究提供获得移植细胞新方法。方法采用健康人脑脊液体外诱导大鼠骨髓间充质干细胞分化为神经样细胞。通过形态学、免疫荧光组化法和Westernblot法对诱导后的细胞进行鉴定。结果运用全骨髓贴壁接种法分离培养的骨髓间充质干细胞生长旺盛、纯度高、细胞长势良好。健康人脑脊液诱导3天后可向神经样细胞分化,可表达神经细胞标志物巢蛋白(NSE)、胶质纤维酸性蛋白(GFAP),细胞长势良好。结论健康人脑脊液可在体外定向诱导大鼠骨髓间充质干细胞分化为神经样细胞。Objective To investigate the differentiation of rat bone marrow mesenchymal stem cells (BMSCs) into neuron like cells in order to provide new techniques for furthel animal experiments. Methods BMSCs isolated from rat bone marrow were differentiated into neuron like cells after induced by human cerebrospinal fluid. The cells were then i- dentified using morphological observation, immunocytochemistry and Western blotting method. Results BMSCs grew well and proliferated quickly with high purity. The cells were induced into neuron like cells three days after exposure to human cerebrospinal fluid. The expression of nestin (NSE) and glia fibrillary acidic protein (GFAP) was determined. Conclusion Human cerebrospinal fluid can induce BMSCs to differentiate into neuron like cells.
