摘 要:目的观察地佐辛对丙泊酚逆行性遗忘作用的影响。方法按分层随机区组设计将72只昆明种小鼠分成6组(n=12):生理盐水组(Ns组)、丙泊酚组(P组,200mg·kg^-1)、地佐辛5mg·kg^-1组(D5组)、地佐辛10mg·kg^-1组(D10组)、丙泊酚+5mg·kg^-1地佐组(PD5组)、丙泊酚+10mg·kg^-1地佐辛组(PD10组)。通过避暗试验、跳台试验测定各组给药后24、48、72h的潜伏时间和错误次数。结果用药24h后,避暗试验中,与NS组相比,P组潜伏期明缩短(P〈0.05);与P组相比,地佐辛单用及两药合用组潜伏期均明显延长(P〈0.05或P〈0.01),错误次数均减少(P〈0.05)。跳台试验中,与NS组比较,P组、D10组潜伏期缩短(P〈0.05),PD10组错误次数增多(P〈0.05),但与P组相比,各组潜伏期和错误次数均无统计学差异。48、72h后,各组数据的差异均无统计学意义。结论大剂量腹腔注射丙泊酚具有逆行性遗忘作用,地佐辛可减轻丙泊酚对小鼠学习记忆的影响。Objective To observe the impacts of different doses of dezocine (D) on the retrograde amnesia induced by propofol (P). Methods A total of 72 Kunming mice were randomly divided into six groups (n = 12) : a normal sa- line group (Group NS), a propofol group (Group P, 200 mg·kg^-1) , a 5 mg·kg^-1 dezocine group (Group D5 ), a 10 mg·kg^-1dezocine groups (Group Dlo ) , and a propofol and 5 mg·kg^-1 dezocine group (Group PD5 ) and a propofol and 10 mg·kg^-1 dezocine group (Group PD10 ). The step -down test and step -through test were conducted to recorde the latency period and number of errors of each group 24 h, 48 h, and 72 h after treatment. Results Twenty - four hours later, compared with group NS, the latency time of Group P was obviously shortened in the step - down test ( P 〈 0.05 ). Compared with Group P, all groups except Group NS showed significantly prolonged latency time (P 〈 0.01 or P 〈 0.05 ), with less errors ( P 〈 0.05 ). In the step - through test, compared with Group NS, the latency time of Groups P and D,0 were reduced ( P 〈 0.05 ) , and the number of errors was increased for Group PD10 ( P 〈 0.05 ). However, compared with Group P, no significant difference was found in latency time and the number of errors. After 48 h and 72 h, no statistical significance was found among these groups. Conclusion A large dose of propofol can cause retrograde amnesia, while its effects on learning and memory in mice can be relieved by administration of dezocine.
