摘  要:目的探讨骨髓源性心肌干细胞来源的外切体对大鼠心肌缺血再灌注损伤及Akt/GSK-3β通路的影响。方法64只SD大鼠随机分为假手术组(S组)、缺血再灌注组(I/R组)、外切体干预组(Ex组)、外切体干预联合PI3K抑制剂组(Ex+L组)。再灌注后2 h各组取8只大鼠,ELISA法测定血清乳酸脱氢酶(LDH)和肌酸激酶同工酶(CK-MB)水平,Western blot测定检测心肌组织Akt和P-GSK-3β水平;其余8只大鼠存活4周后用Masson染色检测心肌组织中胶原纤维。结果与I/R组相比,Ex组血清CK-MB、LDH水平降低,心肌胶原纤维减少,心肌Akt和P-GSK-3β水平增加(均P<0.01)。结论骨髓源性心肌干细胞来源的外切体可减轻大鼠心肌缺血再灌注损伤,这种保护作用可能与Akt/GSK-3β信号通路激活有关。Objective To investigate the effect of exosomes from marrow-derived cardiac stem cells(MCSCs) on myocardial ischemia-reperfusion(I/R) injury and Akt/GSK-3β pathway in rats. Methods Sixty-four SD rats were randomly divided into sham operation(S), I/R, exosome(Ex), and exosome + PI3 K inhibitor(Ex+L) groups. Serum levels of lactate dehydrogenase(LDH) and creatine kinase isoenzyme MB(CK-MB) and myocardial expression of Akt and P-GSK-3β proteins in 8 rats of each group were respectively measured by ELISA and Western blot 2 hours after I/R. Collagen fibers of myocardial tissue in the remaining 8 rats of each group were determined by Masson stain after 4 weeks. Results Compared with I/R group, serum levels of CK-MB and LDH and myocardial proportion of collagen fibers were decreased(P〈0.01), while myocardial expression of Akt and P-GSK-3β was increased in Ex group(P〈0.01). Conclusion MCSCs-derived exosomes can alleviate the myocardial I/R injury, which might be related to the activation of Akt/GSK-3β pathway.
