摘  要:目的制备猪脑神经肽(PBNC)并检测其对H2O2诱导的PC12细胞氧化损伤的保护效果。方法采用碱化-酶解法制备PBNC,高效液相色谱法测定PBNC分子量;利用H2O2诱导PC12细胞产生氧化损伤,MTT法检测细胞活性、流式细胞术检测细胞凋亡及活性氧水平。结果 PBNC中分子量为250-2 000的多肽约占81%。PBNC减轻了H2O2诱导的PC12细胞毒性作用,降低了凋亡率及活性氧生成。结论 PBNC对H2O2诱导的PC12细胞氧化损伤具有保护作用。Objective To prepare the porcine brain-derived neuropeptide compound(PBNC) and investigate its protective effect on H2O2-induced oxidative damage of PC12 cells. Methods PBNC was prepared by basification-enzymolysis method, and its molecular weight(Mr) was determined by HPLC. After the oxidative damage of PC12 cells was induced by H2O2, cell viability, apoptosis and reactive oxygen species(ROS) were determined by MTT and flow cytometry, respectively. ResultsApproximately 81% of PBNC were polypeptides with molecular weights of 250-2000. PBNC reduced the cytotoxicity, apoptosis and ROS production in H2O2-treated PC12 cells. Conclusion PBNC can be neuroprotective for H2O2-induced oxidative damage of PC12 cells.
