摘 要:目的探讨氯喹对脂多糖(LPS)诱导小鼠单核巨噬细胞系RAW264.7细胞损伤的影响。方法建立LPS诱导RAW264.7细胞损伤模型,并以四甲基偶氮唑蓝比色法测定不同浓度LPS和氯喹对RAW264.7细胞活性的影响;碘化丙啶染色法检测细胞坏死率。结果 200 mg/LLPS作为细胞损伤造模质量浓度(P<0.01)。氯喹浓度<5μmol/L时对RAW264.7细胞活性无影响(P>0.05);1.25、2.5μmol/L氯喹预处理分别导致细胞活性增加(P<0.05)、坏死减少(P<0.01),而5μmol/L氯喹促进细胞坏死(P<0.01)。结论一定浓度氯喹对LPS诱导的RAW264.7细胞损伤具有保护作用。Objective To investigate the effect of chloroquine on lipopolysaccharide(LPS)-induced damage of mouse monocyte-macrophage line RAW264.7 cells. Methods The damage model of RAW264.7 cells was induced by LPS. The influence of different concentrations of LPS and chloroquine on activity and necrosis of RAW264.7 cells was determined by MTT and propidium iodide assays, respectively. Results The ideal concentration of LPS was 200 mg/L for establishing the cell damage model(P〈0.01). Less than 5 μmol/L of chloroquine had no effect on the activity of RAW264.7 cell(P〉0.05). Cell activity was increased at 1.25 μmol/L of chloroquine(P〈0.05), while cell necrosis was decreased at 2.5 μmol/L(P〈0.01) but increased at 5 μmol/L of chloroquine(P〈0.01). Conclusion Certain concentrations of chloroquine have protective effects on LPS-induced damage of RAW264.7 cells.
