致倦库蚊卵黄羧肽酶基因与溴氰菊酯抗性关系的研究

摘  要:本文通过对不同抗性株致倦库蚊Culex pipiens quinquefasciatus卵黄羧肽酶基因(Vitellogeniccarboxypeptidase,VCP)表达量的比较,探讨VCP基因与溴氰菊酯抗药性的相关性.通过幼虫生物测定方法,分别得到致倦库蚊敏感株、野外株和抗性株,半数致死浓度分别为0.00057、0.38900和42.26250 mg/L.采用SYBR荧光定量方法,以致倦库蚊核糖体蛋白L8(ribosomal protein L8)基因作为内参,获得VCP基因在致倦库蚊敏感株、野外株和抗性株校正后(拷贝/l×109RPL8)表达量分别为5.80×10 7、4.53×107、3.07×10 7.致倦库蚊敏感株、野外株和抗性株的抗药性从低到高,而VCP基因表达量却是从高到低,推测两者之间存在着负相关关系.进一步推测VCP基因为致倦库蚊抗性相关的新基因,为研究致倦库蚊的抗药性提供了新的方向.This study focused on the correlation between expression level of vitellogenic carboxypeptidase (VCP) gene and resistance to dehamethrin in different Culex pipiens quinquefasciatus strains. Larval bioassays were carried out in accordance with WHO mosquito susceptibility tests. The LCso value was 0. 00057, 0. 38900 and 42. 26250 mg/L respectively. Quantitative analysis showed the transcription expression of VCP gene was 5.80 × 10^7 , 4. 53× 10^7 , 3.07 × 10^7 using ribosomal protein L8 gene as an internal reference. Regression analysis of expression level of VCP gene and LC50 value to insecticides showed a significant correlation between expression level of VCP gene and LC50 value to dehamethrin. The results suggest that VCP gene of Cx. pipiens quinquefasciatus may be involved in deltamethrin resistance, which provides insight into the study of the pesticide resistance related genes of Cx. pipiens quinquefasciatus.
