摘 要:为利用家蝇幼虫抗菌肽粗提物作用于原代FBL-3肿瘤细胞,探究其对原代FBL-3细胞周期的影响,本文通过构建小鼠肿瘤模型,获取原代FBL-3肿瘤细胞,之后用家蝇幼虫抗菌肽粗提物作用于该肿瘤细胞,运用流式细胞术检测每个实验组的细胞增殖周期的分布情况.经家蝇幼虫抗菌肽粗提物作用后的FBL-3原代肿瘤细胞,其细胞增殖周期发生了如下改变:G0/G1期细胞比例升高,S期细胞比例降低,细胞增殖指数降低,与对照组比较具有统计学意义.因此得出结论,家蝇幼虫抗菌肽粗提物可以通过阻滞原代FBL-3肿瘤细胞的增殖周期来诱导细胞凋亡.To investigate the effect of crude extract of antimicrobial peptides in Musca domestica larvae on cell cycle of primary FBL-3 cells. Primary FBL-3 ceils was obtained from the mice tumor model and treated with crude extract of antimicrobial peptides. The cell proliferation cycle of each test group was determined with flow cytometry (FCM) analysis. FCM showed that the crude extract of antimicrobial peptides made the cell cycle of primary FBL-3 changed as follows: the cells in G0/G1 phase increased, in S-phase decreased and the cell proliferation index decreased with statistical significance when compared with that in control group. The effect of antimicrobial peptides crude extract on primary FBL-3 cells showed a dose- and time-dependent manner. It can be concluded that the crude extract of antimicrobial peptides in M. domestica larvae may induce the cell apoptosis by blocking the proliferation of primary FBL-3 tumor cells.
