广西省猪繁殖与呼吸综合征病毒GP2、GP3、GP4蛋白遗传进化分析

摘  要:研究表明,动脉炎病毒GP2、GP3和GP4形成的复合物决定病毒的细胞嗜性,猪繁殖与呼吸综合征病毒(Porcine reproductive and respiratory syndrome virus,PRRSV)GP2和GP4能与细胞受体CD163进行相互作用。GP3和GP4也是刺激猪体产生中和抗体的病毒蛋白。本研究选用本实验室在2013~2014年分离到的11株PRRSV毒株进行ORF2、ORF3和ORF4基因的克隆和序列测定,并与国内外代表毒株进行比对分析。结果显示,11株PRRSV GP2、GP3、GP4核苷酸序列同源性分别为94.6%~99.6%、95.3%~100%、93.9%~100%;与美洲经典毒株VR-2332 GP2、GP3、GP4氨基酸同源性分别为91.4%~93.0%、85.1%~86.7%、89.4%~91.1%;与HP-PRRSV毒株对应氨基酸的同源性分别为96.1%~99.6%、95.3%~98.8%、95.0%~99.4%。序列比对表明:11株PRRSV GP2、GP3、GP4蛋白氨基酸序列未出现缺失和插入,但出现多处点变异。遗传进化分析表明:11株PRRSV均分布在美洲型分支上,且与HP-PRRSV亲缘关系较近。Previous studies have shown that GP2,GP3 and GP4 are the determinants for the cellular tropism of arteriviruses.In Porcine reproductive and respiratory syndrome virus(PRRSV),GP2 and GP4 also interact with the cellular receptor CD163.PRRSV GP3 and GP4 stimulate the production of neutralization antibodies in pigs.Thus,it is important to investigate the genetic variations of GP2,GP3 and GP4.In the present study,ORF2,ORF3 and ORF4 genes of 11 PRRS viruses isolated in 2013- 2014 from Guangxi were amplified,cloned and sequenced for analyzing genetic variations of PRRSV.The results showed that GP2,GP3 and GP4 of these 11 PRRSV strains shared amino acid identities at 94.6%-99.6%,95.3%-100%and 93.9%-100%,respectively.GP2,GP3 and GP4 of these isolates had amino acid identities at 91.4%-93.0%,85.1%-86.7%and 89.4%-91.1%with the prototypical type PRRSV VR-2332 and at 96.l%-99.6%,95.3%-98.8%and 95.0%-99.4%with HP-PRRSV,respectively.Alignment of amino acid sequences revealed no deletion but several mutations in GP2,GP3 and GP4 of these 11 PRRSV strains.The phylogenetic analysis indicated that these 11 PRRSV strains belonged to the North American genotype and were closed to the HP-PRRSV strains.
