摘 要:目的:观察通督调神灸对轻度认知障碍(MCI)大鼠行为学、海马β-淀粉样蛋白(Aβ)表达的影响,探讨通督调神灸抗Aβ过度表达的作用机制。方法:SD大鼠随机分为正常组12只、模型组36只,采用D-半乳糖、亚硝酸钠联合腹腔注射复制MCI大鼠模型。造模成功大鼠随机分为空模组、艾灸组、西药组,每组12只。艾灸组大鼠隔附子饼灸"百会",温和灸"天门""大椎",每次20min,1次/d,治疗2周;西药组大鼠用尼莫地平灌胃,每日3次,共2周。Morris水迷宫测试进行行为学检测,用免疫组化技术检测海马Aβ表达水平,反转录PCR法检测海马早老素-1(PS-1)mRNA、膜酸性蛋白酶-1(BACE-1)mRNA的表达水平,酶联免疫吸附法检测血清白介素(IL)-6含量。结果:空模组大鼠Morris水迷宫逃避潜伏期较正常组明显延长(P<0.01),穿越目标平台次数减少(P<0.01),跨越目标象限占整个游泳时间百分比明显降低(P<0.01),与正常组相比,空模组海马中Aβ、PS-1mRNA、BACE-1mRNA表达及血清IL-6水平明显增高(P<0.01)。与空模组相比,艾灸组、西药组大鼠海马组织Aβ、PS-1mRNA、BACE-1mRNA表达及血清IL-6水平显著下降(P<0.01)。与西药组相比,艾灸组Aβ、PS-1 mRNA、BACE-1 mRNA表达及血清IL-6水平降低(P<0.05)。结论:通督调神灸可能通过降低脑内PS-1mRNA、BACE-1mRNA表达水平,进而阻断Aβ的产生,并通过降低血清IL-6水平,进而阻断Aβ过度表达引发的炎性反应级联效应,以达到治疗MCI的目的。Objective To observe the effect of moxibustion of"Baihui"(GV 20),etc.on learning-memory ability,hippocampal amyloidβ(Aβ)protein expression and immune activity in mild cognitive impairment(MCI)rats,so as to reveal its mechanism underlying improving cognitive impairment.Methods A total of 48 SD rats were randomly divided into normal,model,moxibustion,and medication groups(n=12in each group).The MCI model was established by intraperitoneal injection of2 mL mixture solution containing D-galactose(120 mg·kg-1·d-1)and Sodium Nitrite(90 mg·kg-1·d-1),once daily for40 days.Moxibustion(separated by Radix Aconiti Praeparata cake)was applied to "Baihui"(GV 20),"Fengfu"(GV 16)and"Dazhui"(GV 14)for 20 min,once daily for 2weeks,with one day's rest between two weeks.The rats of the medication group were given with Nimodipine(2mg·kg-1·d-1,t.i.d.)by lavage for 2weeks(except Sundays).The learning-memory ability was detected by Morris maze water swimming tasks.The expression level of hippocampal Aβprotein was detected by immunohistochemistry,and those of hippocampal presenilin-1(PS-1)mRNA and cleaving enzyme(BACE-1)mRNA were detected by real time-PCR,and serum IL-6level was assayed by ELISA.Results Following modeling,the average escape latency of location navigation tests of Morris maze water swimming tests,the expression levels of hippocampal Aβprotein,PS-1mRNA and BACE-1mRNA,and serum IL-6content were significantly increased in the model group(P0.01),while the target-platform crossing times and the percentage of target-quadrant swimming duration of spacial probe trials were remarkably decreased in the model group(P0.01).After moxibustion,the increased escape latency,hippocampal Aβprotein,PS-1mRNA and BACE-1mRNA expression and serum IL-6content,and the decreased target-platform crossing times and the percentage of target-quadrant swimming duration were reversed in both moxibustion and medication groups(P0.01).The effects of the moxibust
