隔核Ghrelin对糖尿病大鼠胃运动的影响

摘　　要：目的探讨隔核Ghrelin对糖尿病大鼠胃运动的调控。方法链脲佐霉素腹腔注射制备糖尿病大鼠模型;荧光免疫组化检测糖尿病大鼠隔核内Ghrelin受体GHS-R1a表达变化;大鼠胃表面固定感应片在体记录胃运动并计算胃运动变化率;并采用中枢注射药物法观察隔核对糖尿病大鼠胃运动的调控作用。所获数据采用方差分析和t检验。结果糖尿病大鼠隔核GHS-R1a表达明显低于正常大鼠(P<0.05)。糖尿病大鼠胃运动明显减弱,胃收缩幅度(P<0.05)和频率(P<0.05)显著降低;隔核注射Ghrelin增强正常和糖尿病大鼠胃运动(P<0.05),且和剂量有关。结论隔核Ghrelin在糖尿病大鼠胃运动调控中发挥重要作用。Objective To study the effect of ghrelin in septum on the gastric motility of diabetic rats. Methods Streptozotocin was injected intraperitoneally to prepare diabetic rats. Fluorescent immunohistochemistry and realtimePCR were used to detect the change of ghrelin receptor GHS-R1a expression. The gastric motility was evaluated by implantation of a force transducer on the surface of rat＇s stomachs and calculated with motility index rate. The effect of which on gastric motility was valued by central drugs injection. analysis of variance and t test.Results The expression of GHS-R1a in septum of diabetic rat was lower than that of normal rats（P〈0.05）. The amplitude and frequency of stomach in DM rats are lower than those in normal rats. 3 the gastric motility of normal and DM rats are increased by injection of ghrelin into septum, and the effect is dose-dependent. Conclusion Ghrelin in septum play an important role in the modulation of gastric motility in DM rat.
